摘要
目的探讨Toll样受体4(TLR4)抑制剂TAK-242对糖尿病及牙周炎小鼠血清中肿瘤坏死因子α(TNF-α)表达及牙龈组织中TNF-α及精氨酸酶1(Arg1)表达的影响。方法建立糖尿病、牙周炎及糖尿病复合牙周炎小鼠模型,并给予TAK-242注射,10 d后取血清运用Elisa方法检测不同小鼠血清中TNF-α的表达;同时获取不同处理组小鼠的牙龈组织,实时荧光定量聚合酶链反应(PCR)检测小鼠牙龈组织中TNF-α及Arg1的表达,数据分析采用独立样本t检验及单因素方差分析。结果糖尿病组、牙周炎组及糖尿病复合牙周炎组小鼠血清中TNF-α及牙龈组织中TNF-α表达较对照组升高,TAK-242的应用可降低血清中TNF-α及牙龈组织中TNF-α表达(F_(血清)=25.329,P_(血清)<0.001;F_(牙龈)=47.901,P_(牙龈)<0.001);而糖尿病组、牙周炎组及糖尿病复合牙周炎组小鼠牙龈组织中Arg1的表达降低,应用TAK-242后Arg1表达上调(F=45.897,P<0.001)。结论 TAK-242可调控炎症相关因子TNF-α及Arg1的表达,可能在牙周炎、糖尿病以及糖尿病复合牙周炎的慢性炎症过程中发挥重要作用。
Objective To investigate the effect of TLR4 inhibitor TAK-242 on the expression of TNF-α in serum and TNF-α and Arg1 mRNA expression in periodontal tissues of diabetic mice and periodontitis mice. Methods Groups of diabetes,periodontitis and periodontitis combined with diabetes mouse models were successfully established and TAK-242 was applied in different groups separately. After 10 days,Elisa method was used to detect the expression of TNF-αin mouse serum and real time PCR was used to detect the mRNA expression of TNF-α and Arg1 in gingival tissue. One-Way ANOVA was applied to analyze the data. Results The expression of TNF-α in mice serum and gingival tissue was increased within diabetic group,periodontitis group and diabetic periodontitis group. The application of TAK-242 could reduce the expression level of TNF-α(Fserum=25.329,Pserum〈0.001;Fgingival tis ue=47.901,Pgingival tis ue〈0.001). The expression of Arg1 in diabetes mellitus,periodontitis group and diabetic periodontitis group decreased in gingival tissues while upregulated after treated with TAK-242(F = 45.897,P〈0.001). Conclusions TAK-242 could regulate the expression of TNF-αand Arg1 and might play an important role in the chronic inflammation of periodontitis,diabetes and diabetic periodontitis.
