摘要
染色体倍性鉴定是开展桑树进化研究及多倍体育种的基础。选择11份只开雄花的雄株桑种质资源,以其有丝分裂中期的造孢细胞为材料,利用去壁低渗法制备染色体标本,观察其染色体数目。按照桑树染色体基数为7(x=7)的理论,在11份桑种质资源中鉴定得到9个四倍体(2n=4x=28)、1个六倍体(2n=6x=42)和1个十八倍体(2n=18x=126)。试验结果表明,造孢细胞是鉴定雄株桑种质资源染色体倍性较为理想的材料。
Identification of chromosome ploidy of mulberry( Morus L.) is the basis of mulberry evolution study and polyploid breeding. In the present study,sporogenous cells at mitotic metaphase of 11 male-flowering mulberry germplasm resources were used as materials to prepare samples using cell wall removal and hypotonic treatment for chromosome ploidy analysis. In accordance with the theory that considers the basic number of mulberry chromosome as 7( x = 7),9of these 11 mulberry germplasm materials were found to be of tetraploid( 2n = 4x = 28),one of hexaploid( 2n = 6x = 42),and one of octadecaploid( 2n = 18 x = 126). The result indicated that sporogenous cell is an ideal material for studying chromosome polyploidy in male mulberry germplasm resources.
