枳砧优系的快速繁殖技术研究 被引量:2

摘要 为将柑橘砧木枳(Poncirus trifoliata(L.)Raf.)的优良单系快速繁殖成遗传背景一致的群体,以大田一号枳优系试管播种实生苗为外植体,进行组织培养快速繁殖研究。结果表明,枳试管播种后暗培养30 d后获得的上胚轴,是较合适的快速繁殖材料。上胚轴节间茎段长度为9 mm、于MT+1 mg/L BA条件下培养时,再生率、再生系数较高。MT+1 mg/L BA有利于上胚轴单芽插条萌芽。MT+0.5 mg/L BA+0.05 mg/L IBA较适合芽增殖,增殖系数可达4.98。1/2 MT+0.1 mg/L NAA+0.1 mg/L IBA+0.6 g/L生物炭的培养条件下,试管苗生根情况最佳,移栽到草炭∶河沙∶锯木屑=1∶2∶1的基质中成活率达79.5%。通过以上流程,可在5个月内实现从种子到植株成活的过程,目前获得129株可供嫁接的枳快速植株。 In order to quickly get sufficient number ol trifoliate orange[ Poncirus trifoliata(h.)Rai.] root-stocks with uniform genetic background, seedlings ol clone Datian No.1 trifoliate grown in vitro were chosen to be propagated by tissue culture.The result indicated that seedlings grown in vitro lor 30 days in the dark were the best explants.The stem segments ol 9 mm in length cultured on MT medium containing 1 mg /L ol BA re-sulted in the highest regeneration rate and index.MT containing 1 mg /L ol BA was the best choice for single-bud cuttings’ germination.The optimal multiplication medium was MT containing 0.5 mg/L BA and 0.05 mg/ L IBA, and the multiplication index was up to 4.98.The root rate ol tissue cultured seedlings maintained 9 5 % while those cultured in 1/2 MT medium containing 0.1 mg/L NAA and 0.1 mg/L IBA and 0.6 g/L charcoal had the best rooting.Then the rooted plantlets were transplanted in turf :sand : sawdust=1:2:1.The survival rate was 79.5%.Up to now,129 in vitro propagated plants could be used for grafting.
