嘌呤受体P2X_7在正常人眼和开角型青光眼患者小梁网细胞上的表达对比研究

摘要 目的探讨嘌呤受体P2X7在体外培养正常人眼小梁网细胞(NTMC)及开角型青光眼(POAG)患者小梁网细胞(PTMC)上的表达差异。方法体外培养、鉴定2种人眼小梁网细胞,利用实时荧光定量PCR和Westernblot方法分别检测2种小梁网细胞上P2X_7受体基因和蛋白的表达差异。结果形态学和免疫学方法成功鉴定了体外培养的2种人眼小梁网细胞;实时荧光定量聚合酶链式反应(PCR)和Westernblot证实正常人和POAG患者小梁网细胞上均有P2X7受体的基因和蛋白表达,且后者表达较前者增高。结论 POAG患者小梁网细胞上的P2X7受体表达较正常人眼小梁网细胞增高。P2X_7受体在小梁网细胞上的作用值得进一步研究。 Objective To compare the expression of purinergic receptor P2X7 in normal human trabecular meshwork cells and POAG patients' trabecular meshwork cells. Methods Trabecular meshwork cells were cultured in vitro and identified by immunohistochemistry. Gene and protein expression of P2X7 were measured by real-time quantitative polymerase chain reaction( real-time q PCR) and western blot analysis. Results Trabecular meshwork cells were successfully identified. Real-time q PCR and western blot analysis confirmed that P2X7 mRNA and protein expressed in both groups. The expression is significantly higher in POAG patients. Conclusions P2X7 receptor expression of PTMC is higher than of NTMC. It's worth for further research to explore the role of the P2X7 receptor in trabecular meshwork cells.
