摘要 目的研究长链非编码RNA MALAT1与非小细胞肺癌患者肿瘤复发和预后的相关性。方法选择原发性非小细胞肺癌患者60例,取其肿瘤组织及癌旁组织,应用RT-PCR技术测定原发性非小细胞肺癌肿瘤组织中MALAT1的表达情况,分析MALAT1表达与非小细胞肺癌患者术后复发及总体生存预后的关系。结果与癌旁组织相比,肺癌组织中MALAT1表达量显著升高(P<0.05)。MALAT1表达与患者年龄、肿瘤直径、肿瘤数量、肿瘤分期、肿瘤分化程度以及术前甲胎蛋白水平等无显著相关性;Cox回归分析结果显示,肿瘤直径>5 cm、有血管侵犯、术前AFP含量≥400 ng/ml以及高MALAT1表达量是影响患者术后复发和总生存的高危因素(P<0.05)。结果 MALAT1在非小细胞肺癌中表达量高与非小细胞肺癌手术后肿瘤复发和预后有显著关系,可为非小细胞肺癌患者的复发和预后提供参考,为临床诊断和治疗提供依据。 Objective To investigate the relationship of long chain non-encoding RNA in the patients with non-small cell lung cancer. Methods 60 cases of primary NSCLC patients accepted were selected, and tumor tissues and the corresponding pa- racarcinoma tissues were collected. The expression of MALAT1 in tissues from these patients was determined by quantitative real-time poly merase chain reaction. The correlation between MALAT1 expression and the recurrence of NSCLC and overall survival prognosis after surgery were analyzed. Results MALAT1 expression in NSCLC was up-regulated, and it was also up-regulated in patients with NSCLC recurrence. Compared with the MALAT1 expression in corresponding paracarcinoma tissues, the MALAT1 expression was obviously higher in NSCLC tissues. The expression of MALAT1 was not significantly correlated with age, tumor di- ameter, tumor number, tumor TNM stage, vascular invasion,pathological differentiation and preoperative AFP content. Cox regression analysis showed that tumor diameter greater than 5 cm, multiple tumor vascular invasion, AFP content which was more than or equal to 400μg/L,and higher expression of MAIATI were the main risk factors affecting recurrence and overall survival of NSCIC patients after surgery(P 〈 0.05). Conclusion The up-expression of MALAT1 in NSCLC could be a promising biomarker to pre-dict the tumor recurrence and prognosis of patients with NSCIC, and it could be used for providing a evidence of the recurrence and prognosis of patients with NSCIC, and a basis for clinical diagnosis and treatment.
