摘要
目的:研究大黄萃取液对心肺复苏后兔肺TNF-α、IL-8水平的影响,探讨大黄萃取液对心肺复苏后兔急性肺损伤的治疗作用及机制。方法:18只日本大白兔,随机分为空白对照组、复苏对照组和实验组,每组各6只,空白对照组、复苏对照组予生理盐水10ml/d灌胃;实验组予大黄萃取液5ml·kg-1·d-1灌胃。分组处理1周后,复苏对照组和实验组大白兔经电刺激诱发室颤制作心肺复苏模型,2h后处死;空白对照组直接处死。取各组血清检测TNF-α、IL-8水平,取兔肺组织用免疫组织化学方法检测TNF-α、IL-8的表达。结果:与空白对照组比较,复苏对照组和实验组血清中TNF-α、IL-8水平明显升高(P<0.05),肺组织中TNF-α、IL-8的表达增多(P<0.05);与复苏对照组比较,实验组血清中TNF-α、IL-8水平明显降低(P<0.05),肺组织中TNF-α、IL-8的表达明显减少(P<0.05)。结论:大黄萃取液可减少心肺复苏后兔血清中TNF-α、IL-8水平,减少肺组织中TNF-α、IL-8的表达,具有肺保护的作用。
Objective:To study changes of the expression of TNF-a and IL-8 caused by rhubarb in rabbits after cardiopulmonary resuscitation,and to investigate the lung protection mechanisms of rhubarb. Method: Eighteen Jap- anese big-ear rabbits were randomly divided into there groups:blank control group, experimental control group and rhubarb group, 6 rabbits/group. All rabbits of blank control group and experimental control group were fed with 0. 9% sodium, 10 ml/d. Rhubarb groups were treated with rhubarb extract 5 ml/kg, d. After one week, rabbits of ex- perimental control group and rhubarb group were produced cardiopulmonary resuscitation model induced by electri cal stimulation of ventricular fibrillation,after 2 h the lever of TNF-α and IL8 in serum were examined by ELISA, and the expressions of TNF-α and IL-8 in lungs were examined by immunohistochemistry. The rabbits of blank control group were directly detected the above indicators. Result:Compared with blank control group, the lever of TNF-α and IL-8 in serum and the expressions of TNF-α and IL-8 in lungs of experimental control group and rhu- barb group were increased significantly (P〈0.05). After treatment with rhubarb, compared with experimental control group,the lever of TNF-a and IL-8 in serum and the expressions of TNF-α and IL-8 in lungs of rhubarb group were decreased significantly (P〈0.05). Conclusion: Rhubarb can decrease the lever of TNF-a and IL-8 in serum and the expressions of TNF-α and IL-8 in lungs,can protect the acute lung injury after cardiopulmonary re- suscitation.
