摘要
目的研究组蛋白甲基转移酶是否参与了巨噬细胞炎症迁移的过程。方法利用斑马鱼尾巴创伤后巨噬细胞向炎症部位迁移的模型,采用全胚胎原位杂交的方法检测巨噬细胞迁移前后58个斑马鱼组蛋白甲基转移酶的表达变化情况。结果斑马鱼尾巴创伤后,巨噬细胞开始向炎症部位迁移,并在尾巴切除后6 h聚集在尾部的巨噬细胞达到最多。原位杂交结果显示在尾巴创伤后巨噬细胞迁移6 h后,有3个斑马鱼组蛋白甲基转移酶的表达水平有变化。结论斑马鱼组蛋白甲基转移酶可能参与了巨噬细胞炎症迁移的过程。
Objective:The aim of this study was to explore whether histone methyltransferases(HMTs)involve in the inflammatory process of macrophages migration.Methods:We detected the expression levels of the zebrafish 58 histone methyltransferases before or post macrophages migration by whole-mount in situ hybridization(WISH), using the model of macrophages migration in response to the tail wound.Results:We show that macrophages migrate rapidly to the site of acute injury induced by tail transection, and at 6 hours-post-tail-transection(6 hptt)the number of macrophages that aggregate in the wound reaches a maximum.The expression level of three histone methyltransferases had changed slightly at 6 hours-post-tail-transection.Conclusion:Some histone methyltransferases may play an essential role in macrophages migration in response to inflammation injury in zebrafish.
