摘要
高效快速的品种(系)DNA指纹图谱鉴定,可为辅助牛鞭草属品种(系)DUS测试和知识产权保护等提供科学依据。本研究从54对SSR引物中筛选出20对引物对牛鞭草属25个DUS测试材料进行鉴定分析。结果表明,20对SSR引物对牛鞭草属25个DUS测试材料共扩增出253条清晰条带,其中多态性条带241条,多态性条带比率(PPB)为95.26%。每对引物的多态信息含量(PIC)为0.599~0.900,平均为0.760。UPGMA聚类分析表明,牛鞭草属25个材料在相似系数为0.668处可分为三大类群。20对SSR引物能鉴别10~22个牛鞭草属材料,引物DHSSR7、引物DHSSR11和引物DHSSR15的有效组合可鉴别所有供试材料。以3对SSR引物扩增的电泳图谱为基础,建立了牛鞭草属25个DUS测试材料的指纹图谱标准模式图,能够快速区分各个供试材料。
Efficient and rapid identification of varieties (or lines) by DNA fingerprinting provides a scientific basis for DUS testing and intellectual property protection of He marthria. In this study, the cultivar (or line) identification among 25 materials of Hemarthria in DUS testing was conducted by using 20 pairs of SSR primers selected from 54 pairs of SSR primers. The results showed that 253 bands in total were amplified by twenty pairs of SSR primers, in which 241 bands were polymorphic bands, and the percentage of polymorphic bands (PPB) was 95.26%. The polymorphic information content (PIC) of each pair of primers ranged from 0.599-0.900, with an average of 0.760. The UPGMA analysis showed that 25 materials of Hemarthria could be divided into three groups at the similarity coefficient of 0.668. 10-22 materials could be completely separated by 20 pairs of SSR primers, and all tested materials could be identified by effective combination of primer pairs of DHSSR7, DHSSR11 and DHSSR15. Based on the electrophoretogram amplified by 3 pairs of primers, the standard DNA fingerprinting for 25 materials of Hemarthria in DUS testing were constructed, and each tested material could be quickly distinguished.
