AMD3100在大鼠短暂性局灶性脑缺血损伤超早期中的抗炎作用

摘要 目的:探讨AMD3100在大鼠短暂性局灶性脑缺血损伤超早期的抗炎作用。方法:将雄性SD大鼠随机分为AMD3100处理组(AMD3100组)、溶剂对照组(PBS组)、假手术组,AMD3100组于再灌注(ischemia-reperfusion,I/R)后给予AMD3100 PBS溶液腹腔注射,假手术组与PBS组于I/R后给予PBS溶液腹腔注射;各组线栓法复制120 min SD大鼠局灶性脑缺血模型,于I/R 24 h后使用ELISA法检测各组缺血区炎症因子IL-10、TNF-α、IL-1β含量。结果:AMD3100组较PBS组IL-10含量高,TNF-α、IL-1β含量低(P<0.05)。结论:AMD3100能够通过抗炎作用对大鼠短暂性局灶性脑缺血损伤起保护作用。 Objective: To explore the anti-inflammatory effect of AMD3100 on the ultra-early time of transient focal brain ischemic injury in rats.Method: Male SD rats were randomly divided into the AMD3100 group, Vehicle group(PBS group) and Sham group, AMD3100 group was given intrapefitoneal injection of AMD3100 after ischemia-reperfusion(I/R), PDB group and Sham group were given of PBS after I/R.The transiem focal brain ischemia model was induced by Zea-longa modified- suture middle cerebral artery occlusion(MCAO).Inflammation factors IL-10, IL-1β, TNF-α were detected by using ELISA method.Result: The level of IL-10 was higher and the levels of IL-1β and TNF- a were lower in AMD 3100 group compared with the PBS group(P〈0.05).Condusion: AMD3100 can improve the prognosis of cerebral ischemia by anti-inflammatory effect.
