摘要
目的:检测广西玉林市药材市场上流通的不同规格等级天麻中天麻素和总天麻素的含量。方法:采用反相高效液相色谱法,色谱柱为Agilent C18液相色谱柱(150mm×4.6mm,5μm),检测波长为220nm,流速为1mL/min,柱温为35℃,流动相为乙腈-0.05%磷酸溶液(3∶97)。结果:天麻素在0.24~4.8μg进样量范围内与峰面积呈良好的线性关系,r=0.999 9,天麻素和总天麻素的平均回收率分别为100.43%、100.69%。24批市售天麻的天麻素含量为0.27%~1.47%,均超过2015版药典中0.25%的标准;总天麻素含量为1.97%~5.50%。结论:市售24批天麻样品中的天麻素含量均符合2015版药典要求;以总天麻素含量为评价指标对天麻进行品质评价的方法较药典以游离天麻素含量为指标更为合理。
Objective：To detect the free gastrodine and total gastrodine in different specifications and grades of Gastrodia elata B1. from Guangxi Yulin drug market. Methods：The determination was carried out with Agilent C18 HPLC column （150mm×4.6mm, 5μm）, using acetonitrile-0.05% phosphoric acid solution （3 ： 97） as mobile phase, at a flow rate of 1mL/min and detected a the wave length 220nm, colunmn temperature 35 ~C. Results： The linear range of gastrodine was 0.24~4.8vg , r= 0. 999 9, and the average recovery rate of free gastrodine and total gastrodine were 100.43 %, 100.69%. The content of free gastrodine in different sample of Gastrodia elata Bl. were 0.27%~0.27^, more than 0.25% of the 2015 edition of pharmacopoeia standards; the content of total gastrodine in different sample of Gastrodia elata B1. Were 1.97%-5.50%. Conclusion： The content of gastrodine in24 market samples conform to the requirements of the 2015 edition of pharmacopoeia; Using the total gastrodine content as evaluation indexes in Gastrodia elata Bl. sample is more reasonable than the pharmacopoeia method to control free gastrodine content of Gastrodia elata Bl. samples for quality evaluation.
