摘要
为研究猪黑色素瘤分化相关基因5(MDA5)对口蹄疫病毒(FMDV)感染宿主细胞的影响,本研究通过构建含有猪MDA5基因的表达质粒,转染细胞后接种FMDV,检测猪MDA5对FMDV的感染能力的影响。结果显示,重组质粒pCMV-Myc-pMDA5能在PK-15细胞中获得表达,并且上调MDA5基因能够一定程度地抑制FMDV感染。本试验结果表明,MDA5具有一定的抗病毒作用,为研究FMDV逃逸宿主天然免疫的机制奠定了一定基础。
To study the effects of Sus scrofa melanoma differentiation related gene 5 (MDAS) on the foot-and-mouth disease virus infection of the host cell, expression plasmid containing the Sus scrofa MDA5 gene was constructed, transfected ceils after vaccination foot-and-mouth disease virus (FMDV) ,to detect the effect of Sus scrofa MDA5 on FMDV infection ability. Results showed that the recombinant plasmid pCMV - Myc - pMDA5 can be expressed in PK 15 cells,and increasing expression of MDA5 genes can inhibit the virus infection in certain extent. The experiment results showed that the MDA5 has certain antiviral effects,which laid a foundation for the research on es cape host innate immune mechanism of foot-and-mouth disease virus
