摘要
为提高奶山羊乳腺上皮细胞泌乳功能基因的表达量,采用分离培养至P_3代的1月龄健康萨能奶山羊乳腺上皮细胞和骨髓间充质干细胞并按2∶1的接种比例混合培养0、24、48、72、96、120、144和168 h(每个处理设置3个重复)后收集细胞,以QPCR检测各时间段SREBP、STAT5、FABP3基因的表达量,比较不同培养时间对奶山羊乳腺上皮细胞SREBP、STAT5、FABP3基因表达量的影响。结果表明:共培养体系下培养时间影响奶山羊乳腺上皮细胞中SREBP、STAT5基因的表达量,却对FABP3基因表达量没有影响。说明共培养体系下奶山羊乳腺上皮细胞与骨髓间充质干细胞混合培养72 h时乳腺上皮细胞泌乳功能基因的表达量最佳。
In order to improve the expression of lactation genes in mammary gland epithelial cells of dairy goats, the mammary gland epithelial cells and bone marrow mesenchymal stem ceils were cultured in a ratio 2:1, the expression of SREBP, STATS and FABP3 were detected by QPCR at 24,48,72,96,120,144,168 h (three replicates per treatment). The results showed that the culture time of co-culture system affected the expression of SREBP and STATS gene in dairy goat mammary gland epithelial cells,but had no effect on FABP3 gene expression. According to the results,the expression of lactation gene in mammary gland epithelial cells after 72 h of mixed culture was the best.
