摘要
在最佳实验条件下,光谱法研究牛血清白蛋白(BSA)和头孢他美酯(CP)结合反应的特征。结果表明:CP对BSA的荧光有猝灭作用,属于静态猝灭。BSA能运输CP,其作用力类型主要为氢键和范德华力。BSA的亚螺旋域ⅡA是主要结合位置,有弱的负协同作用。CP对BSA构象几乎不产生影响,结合位点更接近于酪氨酸。
The interaction between Bovine Serum Albumin (BSA) and Cefetamet Pivoxil (CP) was studied with spectrometry under the optimal conditions. The results showed that the fluorescence of BSA was quenched by CP, which was a static quenching process. BSA could transport CP, which was mainly driven by hydrogen bond and Vander Waals force. The primary binding site for CP was located at sub-domain ⅡA of BSA. There displayed some weakly negative cooperative effect. The conjugation reaction would hardly affect the conforma- tion of BSA, and the binding site was near to tyrosine residue. This test could provid references for its clinical application.
