摘要
旨在建立测定新型中药复方健胃分散片中主要成分延胡索乙素含量的HPLC方法,利用建立的方法测定并比较中药提取机混提法与单提复配法制备的中药复方健胃分散片中延胡索乙素的含量。采用Agilent SB-C18柱(250 mm×4.6 mm,5μm)色谱柱,以甲醇—磷酸盐(65∶35)缓冲液( 值=6.0)为流动相,等度洗脱,于280 nm波长处进行检测。结果表明,延胡索乙素在0.1~1.0μg/m L范围内线性关系良好,r=0.9989,平均回收率为99.99%,RSD=0.91%;中药提取机混提法与单提复配法制备的中药复方健胃分散片中延胡索乙素平均含量分别为2.430 7 mg/g(RSD=0.57%,n=6)和1.937 5 mg/g(RSD=2.17%,n=6)。建立的HPLC方法灵敏度高,操作简便,结果准确,重复性好,可用于中药复方健胃分散片中延胡索乙素含量测定,并且中药提取机混提法所制备的中药复方分散片中延胡索乙素含量较高,效果较好。
The aim of the present study was to establish a high performance liquid chromatography (HPLC) method for determination of tetrahydropalmatine in the Chinese medicine compound Jianwei dispersible tablet, and to evaluate and compare the tetrahydropalmatine content in the dispersible tablet prepared by mixed extraction method and single extraction method using the established HPLC method. The HPLC system was equipped with the Agilent SB-C18 column (250 mm × 4.6 mm, 5 μm); the methanol-phosphate (65:35) buffer (pH value = 6.0) was used as the mobile phase; the isolated gradients were detected at 280 nm wavelength with an ultraviolet detection. The results showed that the established HPLC method had a good linear relationship in the range of 0.1-1.0 μg/mL for tetrahydropalmatine (r=0.9989); the average recovery of tetrahydropalmatine was 99.99% with RSD of 0.91%; the average content of tetrahydropalmatine in the dispersible tablet prepared by mixed extraction method and single extraction method was 2.430 7 mg/g (RSD=0.57%, n=6) and 1.937 5 mg/g (RSD=2.17%, n=6) respectively, indicating that the preparation effect of the former method is better. The results obtained in this study demonstrated that the established HPLC method is sensitive, convenient, accurate and reproducible for determination of tetrahydropalmatine in Chinese medicine compound Jianwei dispersible tablet.
