载脂蛋白AI对单核细胞表型与MCP-1/CCR2的影响

摘要 目的探讨载脂蛋白AI(apoAI)对动脉粥样硬化小鼠血液、脾脏中Ly6C^(hi)单核细胞和单核细胞趋化蛋白1(MCP-1)浓度及CC类趋化因子受体2(CCR2)表达的影响。方法 16只雄性apo E-/-小鼠高脂饲料饲养12周后随机分为对照组和apoAI组,处死前第1、3天分别予以生理盐水和apoAI(40 mg/kg)干预,处死12 h前予以腹腔注射脂多糖(LPS)25μg/只,酶联免疫法检测血浆MCP-1浓度,流式细胞仪检测血液、脾脏中Ly6C^(hi)单核细胞比例。THP-1细胞随机分为对照组和apoAI组,予以相应干预后加入浓度为10μg/L的LPS孵育,Western blotting检测2组中CCR2的表达。结果与对照组比较,apoAI能显著降低血液及脾脏中Ly6C^(hi)单核细胞比例及血浆中MCP-1浓度(P<0.01),体外研究显示apoAI能下调单核细胞CCR2的表达(P<0.05)。结论 apoAI能降低MCP-1浓度及血液、脾脏中Ly6C^(hi)单核细胞比例,抑制CCR2的表达。 Objective To explore effects of apoAI on MCP- 1 levels in the plasma and the Ly6C^hi monocyte proportion in the blood and spleen of atheroselemfie mice, as well as on CCR2 expression in vitro. Methods Sixteen male apoE^-/- mice were fed with high cholesterol diet for 12 weeks. Mice were randomly divided into control and apoAI groups and were administered with PBS or apoAI (40 mg/kg), respectively, via tail vein on the 1st and 3M day before sacrifice. Mice in both groups were administered LPS (25 μg/mouse)via intraperitoneal injection 12 h before sacrifice. Plas- ma levels of MCP- 1 were determined using ELISA, and the Ly6C^hi monocyte proportion was analyzed using flow cytometry. In addition, human monocytic THP-1 cells were randomly divided into control and apoAI ( 10 mg/L) -treated groups, pretreated with corresponding intervention, and incubated with LPS (10μg/L). CCR2 expression levels were measured by Western blotting. Results Compared with the control treatment, apoAI treatment remarkably reduced MCP-1 levels in plasma and Ly6C^hi monocyte proportion in the blood and spleen (P 〈 0.01 ). Furthermore, apoAI treatment inhibited CCR2 expression in monocytes in vitro (P 〈 0.05). Conclusion apoAI can reduce MCP- 1 levels in plasma and the Ly6C^hi monocyte proportion in the blood and spleen and can inhibit CCR2 expression in mouocytes in vitro.
