摘要
目的:检测天然猪链球菌溶血素(n SLY)对人脑微血管内皮细胞凋亡的影响,从而探讨猪链球菌致宿主脑膜炎的发生机制。方法:使用猪链球菌野生株05ZYH33上清、sly基因敲除突变株ΔSLY上清、天然SLY蛋白分别与人脑微血管内皮细胞h CMEC/D3作用4 h后收集细胞,分别用流式细胞术和蛋白免疫印迹技术检测h CMEC/D3细胞凋亡的情况。结果:与对照组比较,天然SLY蛋白可以通过诱导肿瘤坏死因子α(TNF-α)上调显著促进h CMEC/D3细胞发生凋亡,同样含天然SLY的猪链球菌2型野生株05ZYH33上清也通过该途径促进h CMEC/D3细胞凋亡,但突变株ΔSLY上清则不能影响h CMEC/D3细胞的凋亡。结论:猪链球菌溶血素可以通过TNF-α促进人脑微血管内皮细胞发生凋亡,可能是猪链球菌致脑膜炎的发病机制之一。
Objective: To explore the mechanism of meningitis caused by Streptococcus suis, we detect the effect of native S.suis hemolysin(nSLY) on the apoptosis of human brain microvascular endothelial cells.Methods: We used wild strains S.suis 05ZYH33 supernatant, sly knockout mutant strains supernatant and native SLY protein to stimulate the human cerebral microvascular endothelial cells hCMEC/D3 respectively, and collected the cell after 4 h. Furtermore, flow cytometry and protein immunoblot technology were used to observe the apoptosis of the hCMEC/D3.Results: Compared with control group, the natural SLY protein could promote significantly hCMEC apoptosis by inducing the expression of TNF-α. Consisting with the natural SLY protein, the wild strains S.suis 05ZYH33 supernatant also promote hCMEC/D3 apoptosis by the same way, but sly knockout mutant strains supernatant could not affect hCMEC/D3 apoptosis.Conclusion: Suilysin can promote the brain microvascular endothelial cells apoptosis induced by TNF-α, which may be one of the pathogenesis of meningitis caused by S.suis.
