龙眼多聚半乳糖醛酸酶基因DlPG1表达模式与幼果脱落关系研究 被引量:4

摘要 从龙眼中分离获得1个多聚半乳糖醛酸酶(PG)基因,命名为DlPG1。DlPG1基因与多种物种PG基因氨基酸序列有较高的同源性,其中与同属于无患子科的荔枝同源性最高,为95%,且与荔枝的亲缘关系最近。DlPG1基因包含PG基因特有4个保守区中的3个,DlPG1属于E进化支PG基因家族成员。环剥摘叶处理可以显著促进龙眼幼果脱落,对照累积落果率为6.12%,处理为89.85%,相对落果率高峰出现在处理后第5天。处理的离区DlPG1基因表达量始终高于对照,且表达量高峰出现在处理后第4天。基因表达量的高峰早于相对落果率高峰出现,据此推测,DlPG1基因可能是调控龙眼幼果脱落的关键基因。截至目前,DlPG1基因是首例在龙眼中发现的属于E进化支的可能与器官脱落相关的PG基因。 One polygalacturonase (PG) gene named DlPG1 was isolated from longan. Amino acid sequence of DlPG1 showed high homology with the PG genes in other plants. The highest homology was found in litchi (95%). DlPG1 contained three of four typical conserved domains of PG gene. DlPG1 was clustered into clade E, belonging to PG gene family. Girdling plus defoliation could significantly increase the rate of fruitlet abscission. The cumulative fruit abscission of control and treatment were 6.12% and 89.85%, respectively. The peak of relative fruitlet abscission rate (RFAR) occurred in the fifth day after treatment (DAT). The expression of DlPG1 gene at abscission zone of the treatment was higher than that of control. The peak of DlPG1 gene expression appeared in the fourth DAT. The peak time of DlPG1 gene expression in abscission zone was appeared earlier than the RFAR. Thus, DlPG1 gene might he the key gene regulating the fruitlet ab- scission of longan. To our knowledge, it was the first case to report that a PG gene, belonging to the clade E, played a role on organ shedding in longan.
