摘要
以青海云杉4个种源成熟胚为外植体,在4种基本培养基和含有不同激素组合的改良1/2LV培养基中进行体胚诱导,结果表明:改良1/2LV培养基为最适宜基本培养基,以此培养基为基础附加6-BA 2.25 mg·L^(-1)+2,4-D 6.60 mg·L^(-1)对1#、2#、4#种源诱导率均超过30%,分别达到38.9%、33.4%、35.4%,对3#种源诱导率也达到29.6%。在同一水平激素组合处理中,1#、4#种源诱导率高于2#、3#种源,原生种源外植体易于启动并完成脱分化过程,而人工次生种源外植体则不易被诱导。
The somatic embryo induction was carried out in 4 minimal medium and improved 1/2LV medium which con-tained different hormone combination by taking 4 provenance mature embryo of Picea crassifolia as explants. T h e result showed that the improved 1/2LV medium was optimum minimal medium. The inductivity of 1#, 2#, 4# provenance were exceeded 30 % by adding 6 - BA 2. 25 mg·L^-1+ 2,4 - D 6. 60 mg·L^-1 based on improved 1/2LV medium. They were 38.9 % , 33.4 % , 35.4 %. The inductivity of 3# provenance was 29.6 % . The inductivity of 1#, 4# prov enance were higher than 2#, 3# provenance. The native provenance was easy to start and finish the dedifferentiation process. However, artificial secondary provenance explants was not easy to be induced.
