当归挥发油对自发性高血压大鼠肝脏脂代谢的影响 被引量:10

摘要 目的探讨当归挥发油对自发性高血压大鼠肝脏脂代谢的改善作用及其机理。方法将40只8周龄雄性自发性高血压大鼠随机分为模型组,当归挥发油低、中、高剂量组和缬沙坦组,并用同周龄的Wistar大鼠作为正常对照组。分别灌胃4周后,取大鼠肝脏组织,HE染色观察肝脏组织形态学的变化,免疫印迹法检测TRIB1、TRL3、AHSG蛋白水平的表达。结果 HE染色显示,与模型组比较,当归挥发油各组病变分别有不同程度的减轻,缬沙坦组未见明显异常;TRIB1、TRL3、AHSG蛋白在模型组,当归挥发油中、高剂量组表达上调,且呈差异性表达,在当归不同剂量组之间亦呈差异性上调表达。结论当归挥发油对自发性高血压大鼠脂代谢有调节作用,可能通过上调TRIB1、TRL3、AHSG等蛋白的表达量抑制动脉粥样硬化,改善自发性高血压大鼠的脂代谢而达到降压的目的。 Objective To study the influence of essential oil of Angelica sinensis(oliv.)Diels on hepatic lipid metabolism in spontaneously hypertensive rats in order to shed light on the function and mechanism of this oil.Methods Forty eight-week old male spontaneously hypertensive rats were divided randomly into the model group,essential oil of Angelica sinensis(oliv.)Diels groups(low,medium and high)and valsartan group.Wistar rats of the same age were used as the normal control.After four weeks of intragastric administration,the liver tissues of the rats were taken out.HE staining was used to observe the changes of liver tissue morphology while Western blotting was used to test the expressions of TRIB1,TRL3 and AHSG proteins.Results HE staining showed that compared with the model group,lesions in each group of the essential oil of Angelica sinensis were reduced to different degrees.There was no obvious abnormity in valsartan group.Western blotting showed that the expressions of TRIB1,TRL3 and AHSG proteins were up-regulated in the model group and essential oil of Angelica sinensis(oliv.)Diels groups(low and high).There was significant difference in the expressions of these proteins between different dose groups of Angelica sinensis.Conclusion Angelica naphtha can regulate lipid metabolism in spontaneously hypertensive rats by raising the TRIB1,TRL3 and AHSG protein expressions.
