双波长分光光度法测定化橘红中总黄酮的含量 被引量:6

摘要 目的:建立一种测定化橘红中的总黄酮含量时能有效消去样品溶液背景干扰的方法。方法:甲醇超声提取总黄酮;最大吸收波长283nm为测定波长,325nm为参比波长,双波长分光光度法测定总黄酮含量。结果:标准曲线在0~80μg浓度范围内线性关系良好,相关系数r=0.9999;方法稳定性在0~120min良好,RSD%为1.1%;三个浓度水平加样回收率在98%~103%之间。结论:本方法简单易行,适用于化橘红产品的质量控制。 Objective To establish a method for the determination of the total flavonoids content in exocarpium citri grandis and can effectively eliminate the sample solution background interference. Methods The total flavonoids were extracted by ultrasonic method in Methanol. The maximum absorption wavelength was 283nm, the wavelength was 325nm, the content of total flavonoids was determined by standard curve method. Results The calibration curve was linear in the concentration range of 0 -80μg, the correlation coefficient was 0. 9999. The stability of the method was good at 0 - 120min and RSD% was 1.1%. The recoveries of the three levels were 98 - 103% . Conclusion The method is simple and can be used for the quality control of the exocarpium citri grandis.
