摘要
采用乌氏黏度计测定不同pH条件下牛血清白蛋白(BSA)的黏度,运用Wolf方程描述BSA的黏度变化行为.详细讨论了不同pH条件下BSA的特性黏数(intrinsic viscosity,[η])与其构象的关系,并分析了BSA比特性黏数(specific intrinsic viscosity,{η})的组成变化.研究结果表明,采用这种新型的黏度法研究球状蛋白质的构象或溶液性质是科学、可行的.
The viscosities of bovine serum albuminin (BSA) in different pH buffers were measured using Ubbelohde capillary vis-cometer,and the data were evaluated by Wolf equation to describe the composition dependences of BSA. The intrinsic viscosities([η]) of BSA under different pH were correlated with its conformation,and the composition dependences of specific intrinsic viscosi-ties ([ η] ) of BSA were carefully analyzed.Our results indicate that the new viscometric method can be applied in the study of conformarion and solution properties of spherical protein.
