摘要
目的:研究泽泻提取物泽泻醇-B-乙酸酯对人结直肠癌细胞株HCT116生长的影响。方法:采用不同浓度的泽泻醇-B-乙酸酯分别作用于大肠癌HCT116细胞24、48、72小时后,应用MTT(四氮甲基唑蓝)测定细胞生长抑制效应,流式细胞仪(FCM)检测细胞周期变化。结果:泽泻醇-B-乙酸酯能显著抑制HCT116细胞增殖,且在一定的浓度范围内呈时间和浓度依赖性。FCM检测结果显示随着作用时间的延长或者药物剂量的增加,G_0/G_1期的细胞显著增加,使细胞发生G_1期阻滞,进入S期、M期的比例降低,细胞增殖受抑。结论:泽泻醇-B-乙酸酯能以时间和浓度依赖的方式抑制HCT116细胞生长;泽泻醇-B-乙酸酯将细胞阻止在G_0/G_1期,进入S期、M期的比例降低,从而通过影响细胞周期进程来抑制细胞增殖。
Objective:To explore the effects of alisol-B-acetate extracted from ZeXie (rhizoma alismatis) on the growth of cell line HCT116 of human colorectal cancer. Methods:Alisol-B-acetate in different concentrations was acted respectively on colorectal cancer HCT116 cells in 24, 48 and 72 hours. After that, MTT (methyl thiazolyl tetrazolium) assay was used to determine the cell-growth inhibitory effect. What's more, flow cytometry (FCM) was applied to detect the changes of cell cycle. Results:Alisol-B-acetate can significantlYinhibit the proliferation of HCT 116 cells. And in a certain range of concentration, it was in a time and concentration dependent. Moreover, the re-sults of FCM detection showed that the G0/G1 phase cells increased significantly with the prolonging of the action time or the increase of the dose of the drug. The cell arrest occurred in G1 phase, and decreased in entering the S phase and M phase, and cell proliferation was inhibited as well. Conclusion: Alisol-B-acetate can inhibit the HCT116 cell growth in method of time and concentration dependent. Alisol-B-acetate can inhibit cell proliferation by affecting the cell cycle progression through arresting cells in G0/G1 phase and decreasing their entering into S phase and M phase.
