摘要
以精品肝素为原料,研究超声波结合过氧化氢(H_2O_2)制备低分子肝素(2~8 ku)的工艺条件。以超声时间、超声功率、H_2O_2含量和超声温度为因素,采用单因素试验和响应面试验设计,以2~8 ku分子质量部分所占比例为测定指标,确定低分子肝素制备工艺参数的最优组合。研究结果表明,在超声时间76 min、超声功率405W、H_2O_2含量5.8%、超声温度61℃条件下,得到低分子肝素的平均分子质量(3900±50)u,2~8 ku组分含量的平均值为87.35%,低分子肝素的抗-FXa效价为101.16 IU/mg,抗-FIIa效价为31.13 IU/mg,抗-FXa/抗-FIIa值为3.25。
Test with fine heparin as raw material studied on process conditions for preparation of the low molecular weight heparin(LMWH,2-8 ku) by hydrogen peroxide(H2O2) with ultrasonic The research examined ultrasonic time,ultrasonic power,concentration of H2O2,ultrasonic temperature four factors,and used single factor and response surface experimental design to determine the optimal combination of process parameters for LMWH preparation by content of 2-8ku for measuring indicators.The result show that under the conditions of ultrasonic time 76 min,ultrasonic power 405 W,concentration of H2O 25.8%,ultrasonic temperature 61 ℃,low molecular weight heparin obtained an average molecular weight of(3 900±50) u,content of 87.35% for 2-8 ku and titer of 101.16 IU/mg for anti-FXa,titer of 31.13 IU/mg for anti-FIIa,both the ratio of 3.25.
