摘要
目的:采用高效液相色谱法测定不同产地的侧柏叶药材中氨基酸含量,为侧柏叶药材的质量评价提供一定的标准,并探究炮制对侧柏叶中氨基酸含量的影响。方法:采用柱前衍生化HPLC法,应用氨基酸自动分析仪测定样品中的18种氨基酸;以乙腈-水(1∶1)∶4%醋酸钠缓冲盐为流动相,流速为1.2mL/min,梯度洗脱,检测波长为360nm,柱温为27℃。结果:不同产地侧柏叶药材中,均含所检测的16种氨基酸,其中包括6种人体必需氨基酸,不含色氨酸、赖氨酸,以河北产侧柏叶必需氨基酸所占比例为最高,安徽产总氨基酸含量最高。炮制后16种氨基酸含量均降低,且随着炮制程度的加深氨基酸含量明显降低。结论:可以侧柏叶药材中氨基酸的含量对其药材质量进行评价,炮制前后氨基酸含量差别明显,可考虑不经炮制直接入药。
Objective:To Use high performance liquid chromatography (HPLC) to determine and compare the contents of amino acids in platycladus orientalis leaves from different habitats, and study the effects of processsed on contents of them , to provide a ref- erence for the evaluation of the quality of the medicine herbs. Methods:Pre-column derivatization HPLC,ELite-AAK column was used:the mobile phase consisted of acetonitri le-water(1 : 1)-4% sodium acetate buffer,a flow rate was set at 1.2mL/min with gra- dient elution, the wavelength was 360 nm, the column temperature was 27℃. Results:The medicine herbs included all of the detec tion of 18 amino acids,included 8 kinds of essential amino acids from different origins. The contents of the highest essential amino acids was in Hebei and total amino acids was in Anhui. 18 amino acids were decreased after processing, and with the degree of deepe- ning the amino acid contents decreased significantly. Conclusion: The contents of amino acids in the medicinal materials of platycla- dus orientalis leaves has a certain significance to evaluate its quality. Prepared the contents of amino acids before and after being pro cessed the difference was obvious, can be considered without processing directly into the medicine.
