摘要
目的探讨单唾液酸四己糖神经节苷酯对大鼠蛛网膜下腔出血早期脑损伤的影响。方法选择体质量300~350 g SD雄性大鼠60只,按照血管内穿刺法制作大鼠蛛网膜下腔出血模型,将大鼠随机分为治疗组、对照组,每组各30只。治疗组给予单唾液酸四己糖神经节苷酯腹内注射,对照组只进行穿刺手术。所有大鼠于模型建立后24 h予以麻醉后灌流固定,以TUNEL、Neu N法检测大鼠大脑皮质细胞凋亡程度。应用Sugawara出血量评分方法将血肿量分为中、重度出血并分组进行比较。结果治疗组出血程度中、重度大鼠的细胞凋亡比例明显低于对照组,差异有统计学意义(P<0.05)。结论单唾液酸四己糖神经节苷酯具有减少神经细胞凋亡,减轻早期脑损伤的作用。
Objective To investigate the effect of monosialotetrahexosylganglioside on early brain injury( EBI) of moderate and serve level subrachnoid hemorrhage( SAH) in rats. Methods A total of 60 male rats weighed 300-350 g were randomly divided into the treatment group( n = 30) and the control group( n = 30). Rats in the treatment group were given monosialotetrahexosylganglioside intraperitoneal injection,the others in the control group were given piercing operation. All the rats received perfusion fixation after deathly anesthetization 24 hours after model establishment,and the brains were taken for immunofluorescence staining of TUNEL,Neu N to investigate the ratio of neuron apoptosis. Using Sugawara scoring method to measure the volume of hemorrhage and divid the rats into moderate level and severe level,and the comparison was made between the groups. Results The ratio of apoptosis in moderate and severe levels of rats in the treatment group was significantly lower than that in the control group,the difference had statistical significance(P〈0. 05). Conclusion Monosialotetrahexosylganglioside could reduce the ration of nerve cell apoptosis and may alleviate the symptom of early brain injury.
