香水莲花提取物对胰脂肪酶活性的抑制作用 被引量:13

Inhibitory effect of extracts from Nymphaea hybrid flowers on pancreatic lipase

摘要 采用96微孔板法测定香水莲花整花、雄蕊、花瓣、花托醇提物以及雄蕊不同极性部位提取物对胰脂肪酶的抑制活性;通过酶反应动力学方法确定雄蕊乙酸乙酯提取物对胰脂肪酶的抑制类型,并考察其在人工模拟消化肠液中对胰脂肪酶抑制效果的影响。结果表明,香水莲花各部位的醇提物对胰脂肪酶都具有一定的抑制作用,雄蕊醇提物的抑制作用最强。雄蕊不同极性部位提取物对胰脂肪酶的抑制活性由强到弱依次是乙酸乙酯提取物(半数抑制浓度IC_(50)=5.64mg/mL)>二氯甲烷提取物(IC_(50)=8.75 mg/mL)>正己烷提取物(IC_(50)=11.80 mg/mL)>正丁醇提取物(IC_(50)=22.47 mg/mL)。乙酸乙酯提取物对胰脂肪酶的作用类型为非竞争性抑制;经体外模拟消化后,对胰脂肪酶仍有较强的抑制效果,而且抑制率随浓度的升高而增大。因此,香水莲花提取物是一种具有胰脂肪酶抑制活性的优良植物源原料,可用于降脂功能性产品的开发。 Inhibitory activity of ethanol extracts from Nymphaea hybrid flowers containing whole flowers, stamens, petals and tori as well as different polar extracts from stamens was determined by 96 microporosity plates. The inhibition type of ethyl acetate extracts was confirmed by enzymatic kinetics assay. After simulated digesting by artificial intestine fluid, the inhibitory effect of ethyl acetate extracts on pancreatic lipase was measured. The results indicated that each ethanol extracts performed inhibition activity while the stamens extract showed the best. The order sequenced by inhibitory ability on pancreatic lipase from strong to weak was: ethyl acetate extract(IC(50)=5.64 mg/mL)〉methylene chloride extract(IC(50)=8.75 mg/mL)〉hexane extract(IC(50)=11.80 mg/mL)〉butanol extract(IC(50)=22.47 mg/mL). Moreover, the inhibition type of ethyl acetate extracts was non-competitive inhibition. In addition, the digested ethyl acetate extracts still exhibited strong lipase inhibitory activity and its inhibitory rate increased with the rising of concentration. Thus, extracts of Nymphaea hybrid flowers could be used as good plant-originated raw materials with inhibitory effect on pancreatic lipase for the development of lipidlowering functional products.
