甘薯膳食纤维对二甲基肼诱发结直肠癌预防作用的实验研究 被引量:6

Experimental study on the preventive effect of sweet potato dietary fiber on colorectal cancer induced by 1,2-dimethylhydrazine

摘要 目的探讨甘薯膳食纤维对二甲基肼(1,2-dimethylhydrazine,DMH)诱发结直肠癌(colorectal cancer,CRC)的预防作用。方法 90只ICR小鼠随机分为正常对照组,单纯甘薯膳食纤维组,肿瘤模型组,低、中、高剂量甘薯膳食纤维预防组。肿瘤模型组和预防组以DMH 20mg/kg腹腔注射,每周1次,预防组分别喂饲含10%、20%和30%甘薯膳食纤维的饲料。观察动物处理期间的一般表现,第32周处死所有动物,检测血清超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA),计算肿瘤发生率,做组织病理学检查,采用SPSS 17.0软件,进行t检验以及χ2检验,P<0.05为差异有统计学意义。结果实验期末,高剂量预防组小鼠平均体质量(46.7±2.5)g,高于肿瘤模型组(42.3±4.5)g,差异有统计学意义(t=3.076,P=0.007)。高剂量预防组血清SOD水平(179.45±8.42)U/L,高于肿瘤模型组(160.80±22.62)U/L,差异有统计学意义(t=2.957,P=0.007);高剂量预防组肿瘤发生率33.33%,低于肿瘤模型组84.62%,差异有统计学意义(P=0.005);高剂量预防组结直肠黏膜腺体增生程度低于肿瘤模型组。结论甘薯膳食纤维对于DMH诱发CRC具有一定预防作用,其机制可能与抗氧化作用、抑制细胞异常增生、促进肠道致癌物排泄从而降低其对结直肠黏膜的作用效应有关。 Objective To explore the preventive effect of sweet potato dietary fiber on colorectal cancer induced by 1, 2-dimethylhydrazine (DMH) . Methods A total of 90 ICR mice were randomly divided into normal control group, sweet potato dietary fiber group, tumor model group, prevention groups with low, medium and high dose of sweet potato dietary fiber. The mice were injected intraperitoneally with DMH at the dose of 20 mg/kg body weight once a week in the tumor model group and the prevention groups. The mice were fed with food containing 10%, 20% and 30% sweet potato dietary fiber in the prevention groups. In the 32th week, all mice were sacrificed and serum SOD, MDA were measured. The tumor incidence were calculated. The histopathological examinations were performed. The t test and chi-square test were used for data statistical analysis. Results At the end of our experiment, the average body weight (46.7±2.5) g and average serum SOD level (179.45±8.42) U/L in the prevention group with high dose of sweet potato dietary fiber were signifi cantly higher than those in the tumor model group (Pboay weight = 0. 007 ; PSOD = 0. 007) . The tumor incidence (33.33 % ) was significantly lower in the prevention group with high dose of fiber than that in the tumor model group (P= 0. 005) . The degree of glandular hyperplasia in colorectal mucosa in the prevention group with high dose of fiber was significantly lower than that in the tumor model group. Conclusions Sweet potato di etary fiber has a certain preventive effect on colorectal cancer induced by DMH, which may be due to its an tioxidant ability, inhibition of abnormal cell proliferation and enhancement of the excretion of intestinal carcino gens thereby reducing the toxic effect on colorectal mucosa.
