摘要
目的:探讨哌立福新对裸鼠皮下瘤生长的抑制作用。方法:建立荷人食管癌Eca109细胞的裸鼠皮下瘤模型,分别观察对照组及经哌立福新、顺铂和哌立福新联合顺铂治疗后荷瘤裸鼠的肿瘤生长情况,应用TUNEL法观察肿瘤组织中细胞凋亡情况,Western-Blot法观察肿瘤组织中Akt、m TOR蛋白表达。结果:哌立福新和顺铂单独使用均使肿瘤生长速度减慢(P<0.05),二者联合对肿瘤生长抑制效果更明显(P<0.01)。TUNEL法检测发现哌立福新在体内能引起Eca109细胞凋亡(P<0.05),Western-Blot法结果表明哌立福新在体内通过抑制Akt/m TOR信号通路的活性,促进细胞凋亡,从而抑制人食管癌Eca109细胞的生长。结论:哌立福新联合顺铂可抑制裸鼠皮下食管癌移植瘤的生长,其机制可能是通过Akt/m TOR信号通路发挥作用。
Objective To investigate the inhibitory effects of perifosine on the growth of subcutaneously transplanted tumor in nude rats and explore its possible mechanism. Methods The model of human esophageal cancer Eca109 cells transplanted subcutaneous was established in nude rats, and then divided into control group, perifosine group, cisplatin group and perifosine combined cisplatin group, the growth of tumor in all rats were observed.The apoptosis of tumor tissues were detected by TUNEL method, the expression of Akt and mTOR protein were examed by Western blot. Results Perifosine or eisplatin alone treatment could inhibit the growth of tumors( P〈0.05), the inhibitory rate increased significantly in the group of perifosine combined cisplatin (P〈0.01). The results of TUNEL method indicated that perifosine could induce the apoptosis ,of Eca109 ee.lls in vivo(P〈0.05) ,and Western blot results showed that perifasine could inhibit the activity of Akt/mTOR of signaling pathway. Gonclusion Pcrifosine combined eisplatin could inhibit the growth of subcutaneously transplanted tumor in nude rats, which is related to inhibiting Akt/mTOR signaling pathway.
