Characterization of the Major Components in Different Green Tea Dietary Supplements using HPLC-MS/MS Method

摘要 目的:比较不同绿茶膳食补充剂中主要活性成分的含量,为绿茶膳食补充剂产品的质量控制提供依据。方法:运用UPLC-MS/MS方法鉴定绿茶膳食补充剂中的主要成分,HPLC法测定主要成分的含量,最后用主成分分析和层序聚类分析评估绿茶膳食补充剂产品的变异性。结果:12种不同绿茶膳食补充剂中的主要成分为儿茶素、表焙儿茶素、咖啡因、表儿茶素、表焙儿茶素-3-没食子酸酯,儿茶素没食子酸酯、表儿茶素-3-没食子酸酯,其含量差异较大,主成分分析和层序聚类分析结果显示12种产品根据主要成分的含量可分为4个组群。结论:不同绿茶膳食补充剂质量差异较大,高度变异可能导致变量生物学效应,因此有必要使绿茶膳食补充剂标准化,发现更多潜在益处。 Objective To compare the contents of major active component in different green tea dietary supplements,so as to provide basis for product quality control.Methods UPLC-MS/MS and co-elution with standard compounds were employed to identify the major components.An HPLC method was used to quantify the identified components in different supplements.Hierarchical cluster analysis( HCA) and principal component analysis( PCA) were used to evaluate the variability of these products.Results Catechin,epigallocatechin,caffeine,epicatechin,epigallocatechin-3-gallate,gallocatechin gallate,and epicatechin-3-gallate were identified as the major components in 12 different green tea supplements purchased from local store or internet.Quantitative analysis results showed that the contents of these components were highly variable across products.PCA and HCA revealed that the 12 products used in this study could be divided into four groups based on the contents of the major components per daily dose.Conclusion This study suggested that the quality of different green tea supplements are highly variable,which most likely lead to variable biological effects.Standardization of green tea supplement is necessary to derive more consistent potential benefits.
