骨桥蛋白对压力过负荷心衰小鼠的影响 被引量:1

摘要 目的同步观察心肌骨桥蛋白(OTN)、血清氨基末端B型利钠肽前体(NT-proBNP)在过负荷心衰(HF)、过负荷心衰重置于正常负荷小鼠心脏的影响。方法选用雄性昆明种系小鼠C57BL/6野生型小鼠,分为Sham组、过负荷HF组、过负荷心衰重新置于正常负荷(HF-NL)组,观察三组形态学改变、血清NT-proBNP浓度以及OPN mRNA表达。结果光镜下Sham组小鼠心肌细胞结构完整,肌纤维排列整齐;过负荷HF组小鼠心肌纤维间隙增宽,心肌细胞呈不同程度变性及肥大,部分心肌细胞呈局灶性坏死;HF-NL组与过负荷HF组比较,心肌纤维间隙增宽得到改善,心肌细胞变性、肥大以及局灶性坏死变轻;血清NT-proBNP浓度在Sham组、过负荷HF组、HF-NL组分别为(54.10±8.91)mg/L、(459.78±22.79)mg/L(与Sham组比较,P<0.01)、(362.67±43.73)mg/L(与过负荷HF组比较,P<0.01)。OPN mRNA表达在正常小鼠中的表达较弱〔(0.95±0.10)%〕;在过负荷HF组增强〔(3.38±0.12)%,与Sham组比较,P<0.01〕,而在HF-NL组表达减轻〔(1.97±0.14)%,与过负荷HF组比较,P<0.01〕。结论OPN参与过负荷心衰置于正常负荷后的心肌重塑过程,抑制OPN表达可能是减轻、减缓过负荷HF发生、发展的干预靶点之一。
