摘要
目的:通过观察活络效灵丹对啤酒诱发痛风模型大鼠的影响,对活络效灵丹治疗痛风的疗效及机制进行探索。方法:将60只雄性SD大鼠随机分为空白对照组,造模啤酒组,加味活络效灵丹高、中、低剂量组,每组12只。除空白对照组外,其余各组大鼠注射尿酸钠诱导痛风模型。空白对照组给予生理盐水灌胃,其余各组均给予啤酒灌胃,加味活络效灵丹高、中、低剂量组同时予以加味活络效灵丹汤液高、中、低剂量灌胃。观察各组大鼠的饮食、二便、毛色、精神状态及活动情况,受试关节周径变化;ELISA法检测关节组织中白细胞介素-1β(IL-1β)的水平。结果:与造模啤酒组比较,加味活络效灵丹高、中、低各剂量组大鼠的饮食、二便、毛色、精神状态及活动情况均有所改善。与空白对照组比较,造模啤酒组大鼠的受试关节周径有所增加(P<0.05);与造模啤酒组比较,加味活络效灵丹高、中、低各剂量组大鼠的受试关节周径均有所减少(P<0.05)。与空白对照组比较,造模啤酒组大鼠的关节组织中IL-1β的表达有所增加(P<0.05);与造模啤酒组比较,加味活络效灵丹高、中、低各剂量组大鼠的关节组织中IL-1β的表达均有所降低(P<0.05)。结论:加味活络效灵丹可通过降低关节组织中IL-1β的表达而起到治疗痛风的作用。
Objective:To probe the therapeutic mechanism of Huoluo Xiaoling Dan ( 活络效灵丹 ) by observing its effect on beer-induced gout rat models.Methods:Sixty male SD rats were randomly divided into a blank control group,a beer model group,and high-dose,medium-dose and low-dose groups of Huoluo Xiaoling Dan, 12 rats in each group.Except the blank control group,the rats in the other groups were induced by sodium urate injection to establish gout models.The blank control group was given intragastric administration of normal saline,while the other groups were given intragastric administration of beer.At the same time,the high-dose,medium-dose and low-dose groups were also given intragastric administration of corresponding doses of modified Huoluo Xiaoling Dan.An observation was made on the rats' diet,urine,stool,hair color, mental state,activity and the circumference of subject joints.ELISA method was used to detect the level of IL-1β in the joint tissue.Results:Compared with the beer model group,the diet,urine,stool,hair color, mental state and activity of rats in the three groups of modified Huoluo Xiaoling Dan were improved to a certain degree.Compared with the blank control group,the joint circumference of rats in the beer model group increased to a certain degree ( P 〈 0.05 ) .Compared with the beer model group,the joint circumference of rats in the high-dose,medium-dose and low-dose groups decreased to a certain degree ( P 〈 0.05 ) .Compared with the blank comrol group,the expression of IL-1β in the beer model group increased to a certain degree (P 〈 0.05 ) .Compared with the beer model group,the expression of IL-1β in the high-dose,medium-dose and low-dose groups decreased to a certain degree ( P 〈 0.05 ). Conclusion:Modified Huoluo Xiaoling Dan treats gout by reducing the exoression of IL-1β in the Joint tissue.
