摘要
目的:研究督脉电针在慢性脑缺血时大鼠脑食欲素受体-2(OX2R)及其配体在不同时间点缺血后的变化。方法:实验中运用分次结扎双侧颈总动脉法建立慢性脑缺血模型,将Wistar大鼠分4组,造模成功后给予督脉电针干预,免疫组织化学法观察OX2R的表达,同时运用ELISA技术检测各组大鼠额叶皮质区Orexin-A和Orexin-B的总含量变化。结果:1个月、2个月时,模型组OX2R及其配体表达高于正常组,同时呈递增趋势;在相同时间进行组间比较,1个月时,模型组和针刺治疗组OX2R及其配体的表达无显著差别,立即针刺组OX2R及其配体的表达较模型组有升高;2个月时针刺治疗组OX2R及其配体表达较模型组有明显升高,而立即针刺组与针刺治疗组相比OX2R及其配体表达有升高。结论:实验观察到OX2R及其配体的表达在1个月和2个月时随着时间延长而升高,经过督脉电针干预后促进了OX2R及其配体表达,同时观察到早期干预对OX2R及其配体表达的影响优于针刺治疗组。
Objective:To explore the effect of electronic acupuncture of DU on OX2R and its ligand expression of rats with chronic cerebral ischemia in different time points. Methods:Establish the cerebral ischemic models by fractionally ligaring bilateral carotid arteries. The Wistar rats were divided into four groups. After the success of building, the electronic acupuncture of DU was used for intervention and measure the OX2R expression by immunohistoehemical technique. Orexin - A and Orexin - B in frontal cortex were examined via ELISA techniques. Results : After one month, the expres- sion levels of OX2R and its ligand were higher and the level of OX2R and its ligand were significantly higher in the mod- el group than those in control group. Compared between groups at the same time, there was no significant difference in statistics of one month between the model group and the acupuncture group, but the expression level of OX2R and its ligand in the immediate acupuncture group were higher than those in the model group. The OX2R and its ligand expression after two months in acupuncture group were increased obviously compared with those in the model group, and the OX2R and its ligand expression of immediate acupuncture group were increased compared with thos in the acupuncture group. Conclusion:The expression levels of OX2R and its ligand have an increased trend with the extension of time during one month and two months. The electronic acupuncture of DU can promote the expression levels of OX2R and its lig- and, and the early intervention is superior to the acupuncture treatment group.
