Application of Supersaturated System in Transdermal Drug Delivery System

摘要 超饱和体系通过提高药物在溶剂中的热力学活度,从而增加药物释放及透皮驱动力,达到促渗目的。与传统物理及化学促渗法相比,该技术具有促渗效果显著、皮肤刺激性小、成本低廉、应用方便等显著优势,可单独使用或与其他促渗手段联用。但超饱和体系属热力学不稳定系统,极易形成结晶,影响透皮效果,在产品制备过程中应通过加入结晶抑制剂等方法维持体系的稳定性。本文简要概述了超饱和体系在经皮给药领域的应用及其结晶抑制现状。 Supersaturated system has been used to enhance the transdermal permeation of drugs effectively by increasing drug thermodynamic activities and thereby the driving force for transiting out of the dosage form and penetrating the stratum corneum. It shows superiorities such as significant enhancement, low skin irritation, low cost and convenient application, compared with other traditional chemical and physical permeation enhancement approaches. This technology can be applied alone or combined with other enhancement methods. The main limitation of supersaturated system is related to its thermodynamic instability leading to drug crystallization, which will affect the skin permeation of drugs. Adding crystal growth inhibitor during the preparation process can maintain the stability of supersaturated system. This review provides a brief overview of the application of supersaturation in transdermal delivery system and its crystal inhibition.
