摘要
目的:观察益肾活血丸改善子宫内膜容受性的情况以及该药治疗本病的作用机理。方法:试验组25例,选取10例做自身前后对照;对照组23例,选取8例做自身前后对照。2组自身对照患者分别于治疗前及治疗后排卵后7~10天刮取少量子宫内膜,余患者仅于治疗后排卵后7~8天刮取内膜,用半定量RT-PCR技术测定子宫内膜中HOXA10、HOXA11、Wnt7a的表达量,并对HOXA10、HOXA11与Wnt7a的关系进行相关性研究。结果:经过3个月经周期的治疗后,试验组、对照组中行自身前后对照患者子宫内膜中HOXA10 mRNA、HOXA11 mRNA、Wnt7a mRNA表达量均较治疗前明显升高,差异均有统计学意义(P<0.05)。经过3个月经周期的治疗后,试验组子宫内膜中HOXA10 mRNA、HOXA11 mRNA表达量稍高于对照组,差异无统计学意义;试验组子宫内膜中Wnt7a表达量明显高于对照组,差异有统计学意义(P<0.05)。HOXA10 mRNA与Wnt7a mRNA在子宫内膜容受性建立过程中呈高度正相关,相关系数r=0.842,P=0.000<0.01,提示二者存在明显的直线相关关系。HOXA11 mRNA与Wnt7a mRNA在子宫内膜容受性建立过程中呈高度正相关,相关系数r=0.864,P=0.000<0.01,提示二者存在明显的直线相关关系。结论:HOXA11可能与HOXA10一样参与了子宫内膜容受性的建立。中成药益肾活血丸可能通过经典Wnt/β-catenin信号通路改善肾虚血瘀型多囊卵巢综合征(PCOS)患者子宫内膜容受性。
Objective：To observe improvement of endometrial receptivity by Yishen Huoxue pill and study its mechanism.Methods：In the 25 patients of the experiment group,selected ten of them as self- control before and after treatment.In the23 patients of the control group,selected eight of them as self-control before and after treatment.The self-control patients in the two groups were scraped a small amount of endometrium after ovulation for 7 to 10 days respectively,before and after treatment.The others were scraped endometrium after ovulation for 7 to 8 days only after treatment.Tested expression of homeobox gene A10（HOXA10）,homeobox gene A11（HOXA11）,Wnt7 a in endometrium with semi-quantitative Reverse Transcripase Polymerase Chain Reaction（RT- PCR）,and studied the correlation among HOXA10,HOXA11 and Wnt7 a.Results：After three menstrual cycle of treatment,expression of HOXA10 mRNA,HOXA11 mRNA,Wnt7 a mRNA in endometrium of the self- control patients of the experiment group and control group were all higher than that before treatment（P〈0.05）.After three menstrual cycle of treatment,expression of HOXA10 mRNA,HOXA11 mRNA in endometrium of the experiment group were slightly higher than those of the control group,the difference being no significance（P〈0.05）.Expression of Wnt7 a in endometrium of the experiment group was higher than that of the control group obviously（P〈0.05）.During the process of establishing endometrial receptivity,HOXA10 mRNA and Wnt7 a mRNA showed a high positive correlation with correlation coefficient r=0.842,P=0.0000.01,which suggested there are obvious linear correlation between the two.There was also a high positive correlation between HOXA11 mRNA and Wnt7 a mRNA during the process,with correlation coefficient r=0.864,P=0.000 0.01,which suggested there are obvious linear correlation between the two.Conclusion：HOXA11 may participate in the establishment of endometrial receptivity the same as HOXA10 does.Chinese patent drag Yishen Huoxue pill can improve endometrial re
