尤瑞克林对脑缺血再灌注模型大鼠缺血侧皮质区内皮抑素表达的影响 被引量:1

Effect of urinary kallidinogenase on the expression of endostatin in ischemic cortex of the rat models after cerebral ischemia reperfusion

摘要 目的观察尤瑞克林对脑缺血再灌注模型大鼠缺血侧皮质区内皮抑素(ES)表达的影响。方法将SD大鼠随机分为假手术组、模型组和实验组3组。模型组和实验组大鼠用线栓法制作大脑中动脉阻塞(MCAO)脑缺血再灌注模型,假手术组接受相似手术处理,但不插入线栓。实验组于再灌注后5 min经尾静脉注射用灭菌生理盐水稀释的尤瑞克林3.5×10^(-3)PNAU/kg,给药体积1 m L/kg;假手术组和模型组经尾静脉注射灭菌生理盐水。于再灌注48 h后进行神经功能缺损评分,采用免疫组织化学法和Western Blot法检测缺血侧皮质区微血管内皮细胞ES的表达。结果与模型组比较,实验组神经功能缺损评分明显降低,缺血侧皮质区微血管内皮细胞ES表达明显下降,差异均有统计意义(P<0.05)。结论尤瑞克林可通过下调缺血侧皮质区微血管内皮细胞ES表达来促进脑缺血再灌注后的血管新生,从而发挥神经保护作用。 Objective To study the effect of urinary kallidinogenase on the expression of endostatin( ES) of microvascular endothelial cell after focal cerebral ischemia reperfusion in rats. Methods Rats were randomly divided into three groups: sham operation group,model group and test group. The middle cerebral artery occlusion( MACO) reperfusion model was made by the suture method. The rats in the test group were injected intravenously with urinary kallidinogenase 3.5×10-3PNAU / kg after making a model for 5 minutes,volume of administration was 1m L / kg; the rats in the sham group and model group were injected with the same dosage of normal saline. The scores of neurological deficit were estimated and the expression of ES was detected by immunohistochemistry and Western blot after reperfusion for 48 hours. Results Compared with the model group,scores of neurological deficit reduced obviously and the ES expression was down-regulated in rats of the test group,the difference was significant( P 〈 0.05).Conclusion Urinary kallidinogenase can induce angiogenesis by down-regulating the ES expression of microvascular endothelial cell,thus play a role of neuroprotection.
