调节小分子GTP结合蛋白Rab27表达对人膀胱癌细胞Cyclin、p-FAK、p-IκB表达的影响 被引量:2

摘要 目的观察调节小分子GTP结合蛋白Rab27表达对人膀胱癌细胞中细胞周期蛋白(Cyclin)、磷酸化黏着斑激酶抗体(p-FAK)、核因子κB抑制蛋白(p-IκB)表达的影响。方法取对数生长期人膀胱癌细胞系BIU-87(Rab27低表达细胞系)用Rab27质粒转染,人膀胱癌细胞系5637(Rab27高表达细胞系)用Rab27干扰质粒转染。采用Western blotting法检测转染前后BIU-87及5637细胞Rab27、Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB。结果与Rab27质粒转染前比较,转染后BIU-87细胞Rab27的相对表达量升高,细胞Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB相对表达量均升高(P均<0.05)。与Rab27干扰质粒转染前比较,5637细胞Rab27的相对表达量降低(P均<0.05),细胞Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB相对表达量均降低(P均<0.05)。结论转染Rab27质粒的BIU-87细胞Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB的表达升高。转染Rab27干扰质粒的5637细胞Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB的表达降低。Rab27可能通过调节NF-κB、FAK信号通路的相关蛋白表达来影响膀胱癌细胞周期。 Objective To observe the effect of regulating the expression of small molecule GTP binding protein Rab27 on the cell cycle protein( Cyclin),phosphorylated focal adhesion kinase antibody( p-FAK) and nuclear factor κB( pIκB) of human bladder cancer cells. Methods Rab27 plasmid and Rab27 siRNA were transfected into BIU-87 and 5637 cells in logarithmic phase,respectively. The levels of Rab27,Cyclin D1,Cyclin E,p-FAK( Tyr925),p-FAK( Tyr576 /577) and p-IκB were analyzed by Western blotting in the bladder cancer BIU-87 and 5637 cells before and after transfection. Results Compared with the Rab27 plasmid transfected BIU-87 cells before transfection,the relative expression of Rab27 was increased,and the expression of Cyclin D1,Cyclin E,p-FAK( Tyr925),p-FAK( Tyr576 /577) and p-IκB was increased( all P 〈 0. 05). After the Rab27 interference plasmid was transfected into 5637 cells,the relative expression of Rab27 was decreased( all P 〈 0. 05),and Cyclin D1,Cyclin E,p-FAK( Tyr925),p-FAK( Tyr576 /577) and p-IκB relative expression was decreased( all P 〈 0. 05). Conclusion After the Rab27 plasmid is transfected into BIU-87 cells,the expression of Cyclin D1,Cyclin E,p-FAK( Tyr925),p-FAK( Tyr576 /577) and p-IκB is increased. After the Rab27 interference plasmid is transfected into 5637 cells,the expression of Cyclin D1,Cyclin E,p-FAK( Tyr925),p-FAK( Tyr576 /577) and p-IκB is decreased. Rab27 may affect the cell cycle of bladder cancer by regulating the expression of NF-κB and FAK signaling pathway.
