CCN3在胶质瘤细胞中的表达及对肿瘤侵袭作用影响的初步研究 被引量:3

摘要 目的以低恶性胶质瘤细胞A172和高恶性胶质瘤细胞U87为研究模型,观察分泌型蛋白肾母细胞瘤过表达基因(NOV/CCN3)在胶质瘤细胞中的表达并初步探讨可能的分子生物学机制。方法用实时荧光定量聚合酶链式反应(qRT-PCR)实验检测A172和U87细胞中CCN3的表达情况,同时用酶联免疫吸附试验(ELISA)检测A172和U87细胞培养上清液中CCN3的蛋白含量,Transwell试验观察CCN3刺激前后A172细胞及U87细胞的侵袭能力。结果 U87细胞中CCN3在mRNA的表达水平是A172细胞中的15.63倍,CCN3在A172细胞培养上清液中的含量是48.62 pg/m L,而在U87细胞培养上清液中的含量是549 pg/m L;U87细胞的侵袭能力强于A172细胞,10 ng/m L的CCN3能明显增强A172细胞的侵袭能力,而对细胞增殖能力无明显影响。结论 CCN3的表达可能与胶质瘤的恶性程度密切相关,并与胶质瘤侵袭能力有关,与肿瘤增殖能力无关。 Objective The expression of secretary protein nephroblastomaoverexpressed gene (NOV/CCN3) in A172 and U87 gloom cell lines with different malignancy was studied and then the function of CCN3 in glioma cells and the possible molecular mechanism were discussed.MethodsQuantificational realtime polymerase chain reaction (qRTPCR) and enzymelinked immunosorbent assay (ELISA) were used to compare the expression of CCN3 between A172 and U87 cells and their culture media. Human recombinant protein CCN3 was used to treat A172 cells, then MTS assay was used to determine the proliferation and transwell assay was used to determine the invasion of A172 cells after treatment with CCN3.ResultsThe expression of CCN3 in mRNA level in A172 cells was 15.63 folds lower than that of U87 cells. CCN3 protein in A172 cell culture media was 48.62 pg/mL compared to 549 pg/mL in U87 cell culture media; U87 cells was more invasive than A172 cells and 10 ng/mL CCN3 promoted A172 invasion with no effect on the proliferation.ConclusionCCN3 expression may be closely associated to the malignancy of glioma and have a relationship with invasion of glioma, but it has no effect on the proliferation.
