摘要
目的 研究脑膜瘤组织中端粒酶的活性及其相关基因端粒重复结合因子1、2(telomererepeat binding factor 1、2, TRF1、TRF2)、端粒酶相关蛋白1(telomerase associated protein 1, TP1)、端粒酶逆转录酶亚基(telomerase reverse transcriptase subunit, TRT)的表达,为临床诊断和治疗开拓新思路。方法 用改良端粒重复序列扩增(TRAP)法检测40例脑膜瘤的端粒酶活性,用免疫组化法检测TRF1、TRF2、TP1、TRT蛋白的表达。结果 脑膜瘤组织中端粒酶阳性率为42.5%,端粒酶阳性率和活性水平随肿瘤恶性度增加而升高。TP1、TRT表达水平和肿瘤恶性度呈正相关。TRF1、TRT2表达水平和肿瘤恶性度呈负相关。结论 端粒酶活性及TRT表达与脑膜瘤恶性度密切相关,TRT是端粒酶活性水平调节的关键。
Objective To investigate the activity of telomerase in meningiomas, expressions of its relevant genes, including telomere repeat binding factor 1(TRF1) , telomere repeat binding factor 2(TRF2) , telomerase associated protein 1(TP1) and telomerase reverse transcriptase subunit (TRT) , for exploring new strategies for clinical diagnosis and treatment of meningioma. Methods The activity of telomerase was detected by modified TRAP method and the expressions of relevant genes of telomerase (TRF1, TRF2, TP1, TRT) were measured by immunohistochemistry method in 40 meningiomas respectively. Results The positive rate of telomerase was 42.5% in meningiomas. The positive rate and activity of telomerase increased with increase of the tumor malignancy degree. The expressions of TP1 and TRT were correlated positively with the malignancy degree of tumor, while the expressions of TRF1 and TRF2 were correlated negatively with the malignancy degree of tumor. Conclusion The telomerase activity and TRT expression are significantly correlated to the malignancy degree of meningioma. The TRT may play a key role in the regulation of telomerase activity.
