不同提取液配方对水稻种子代谢组学研究的影响 被引量:3

摘要 评估不同提取液配方对水稻种子代谢组学研究的影响,并讨论提取液配方选择策略。选择植物代谢组学研究中最常见的3种提取液配方,对扬稻6号及武运粳7号两种水稻成熟种子样品进行非靶标的代谢轮廓分析,超高效液相色谱质谱联用在水稻种子样品中共鉴定出165种已知代谢物(初级代谢产物为122,次生代谢产物为43种)。统计分析表明:不同提取液配方显著影响水稻种子代谢物的检出覆盖率及峰强度:提取液配方3效果最好,但检测和分析工作量均加倍,适合对广度精度要求高的水稻种子代谢组学研究;提取液配方2在不增加工作量的前提下可获得可观的覆盖率和峰强度,效率高,适合一般要求的水稻种子代谢组学研究。 Effects of different extraction methods on rice seed metabolic profiling were evaluated and selection strategy of these methods was discussed.Three commonly used extraction methods were chosen to conduct metabolic profiling analysis of two mature rice cultivars'seeds(Yangdao No.6and Wuyunjing No.7).Ultra-performance liquid chromatography and mass spectrometry(UHPLCMS/MS)was used for detecting metabolites in samples,and then,165 known metabolites were identified.Statistic analysis shows that different extraction methods can remarkably affect the coverage of detection as well as the peak intensity of metabolite.The third extraction method performs best,but doubles the workload of detection and analysis,which is suitable for high-precision and widely studies on metabolomics.The second method can acquire considerable coverage,peak intensity and efficiency,which is suitable for common studies on metabolomics.
