摘要
目的应用UFLC-Q-TOF/MS对黄芪-丹参药对中的化学成分进行鉴定。方法采用Thermo BDS C18(3μm,150mm×2.1mm)色谱柱,以0.1%甲酸水-0.1%乙腈为流动相进行梯度洗脱,流速为0.3mL/min,柱温40℃,进样量5μL。离子源为电喷雾离子源(ESI),负离子模式下采用飞行时间质谱采集数据,全扫描质量范围为m/z 100~1 500。依据保留时间、精确分子量、二级质谱裂解碎片,结合对照品比对初步鉴定化合物。结果从黄芪-丹参药对中初步鉴定或推断出28个化合物,其中丹参中15个化合物,黄芪中11个化合物,另有2个未知化合物。包括皂苷类、有机酸类、黄酮类和醌类化合物。结论该UFLC-Q-TOF/MS可全面、快速分析黄芪-丹参药对中的化学成分,研究结果为对其进行活性成分研究和质量控制提供了一定参考。
OBJECTIVE To identify the main chemical components in herbal pair:Astragali Radix and Salviae Miltior- rhizae by UFLC-Q-TOF/MS. METHODS Thermo BDS C18 (3μm, 150 mm × 2.1 mm) column was employed for the separa- tion with acetonetrile-water (0.1 % formic acid) as mobile phase using a gradient elution program. The flow rate was set as 0. 3 mL/min, the temperature of column was 40 ℃ with injection volume of 5 μL. Electrospray ionization (ESI)-Q-TOF/MS in negative ion mode was performed to obtain the MS spectra with full mass scan m/z ranged from 100 to 1 500. Chemicals were identified based on the retention time, high-resolution mass information, MS/MS fragmentation behaviors and standards. RE- SULTS 28 components were identified or tentatively characterized in herbal pair:Astragali Radix and Salviae Miltiorrhizae, including 15 components in Salviae Miltiorrhizae, 11 compounds in Astragali Radix, and 2 unknown compounds. And these chemicals could be classified as triterpenes, organic acids, flavonoids and quinines. CONCLUSION This method can rapidly and accurately identify the main compounds in herbal pair:Astragali Radix and Salviae Miltiorrhizae, which can help us for the quality control and further phytochemical studies of Astragali Radix and Salviae Miltiorrhizae. in traditional Chinese medicine.
