胰岛素通过Erk通路促进人乳腺癌细胞MCF-7增殖和迁移

摘要 目的观察胰岛素对人乳腺癌细胞MCF-7中Erk信号通路的影响及其对MCF-7细胞增殖、侵袭和迁移的影响。方法分别采用0、50、100和200n M不同剂量的胰岛素处理MCF7细胞0、24、48和72h,采用Western blot方法检测MCF-7细胞中Erk信号通路的磷酸化活化情况,采用MTT方法检测MCF-7细胞的增殖情况。通过伤口愈合实验和Boyden小室检测200n M胰岛素处理72h对MCF-7细胞侵袭和迁移能力的影响。采用Erk信号通路抑制剂预处理MCF-7细胞,观察Erk信号通路在胰岛素促MCF-7细胞增殖、迁移和侵袭能力中的作用。结果同0nM胰岛素处理组相比,不同剂量的胰岛素均可显著上调Erk的磷酸化水平,加强MCF-7细胞的增殖能力(P<0.05),且这种作用具有计量依赖性。同对照组相比,200nM胰岛素处理72h后,MCF-7细胞侵袭和迁移能力均显著增强(t=188.000、95.640,P<0.05)。采用Erk信号通路阻断剂U0126阻断MCF-7细胞中的Erk信号通路后,胰岛素促MCF-7细胞增殖、迁移和侵袭的能力显著降低(P<0.05)。结论胰岛素可通过激活MCF-7细胞中的Erk信号通路,增强肿瘤细胞的增殖、迁移和侵袭能力。 Objective To observe the effect of insulin on Erk signaling pathway and its regulation of proliferation, invasion and migration in breast cancer cells MCF-7. Methods MCF-7 cells were treated with insulin with different concentrations of 0, 50, 100 and 200 nM or in a time dependent manner of 0, 24, 48 and 72 h, then the phosphorylation levels of Erk were detected by Western blot, and the proliferation abilities of MCF-7 cells were detected by MTT. The effect of MCF-7 cells treated with 200 nM insulin by 72 h on the invasion and migration of MCF-7 cells were tested by the wound healing assay and Boyden chamber test. MCF-7 cells were pre-treated with Erk signaling pathway inhibitor to observe the effect of Erk signaling pathway of insulin induced proliferation, invasion and migration. Results Compared with 0nM insulin group, the 50, 100 and 200 nM insulin groups could make active Erk signaling pathway and promote proliferation inMCF-7 cells(P〈0.05), and those effect had concentration dependent. After treated with 200 nM insulin by 72 h, the invasion and migration abilities of MCF-7 cells were significantly increased(P〈0.05). After being blocked Erk signaling pathway by U0126, the abilities of proliferation, invasion and migration of MCF-7 cells treated with insulin were significantly decreased(P〈0.05). Conclusion Through actives Erk signaling pathway, insulin can promote proliferation, invasion and migration of MCF-7 cells.
