摘要
目的探讨褪黑素(melatonin,MLT)对成人外周血CD4+T细胞活化的影响。方法采集健康成年人外周血10mL(n=27),分离、获取单个核细胞,每份标本分成2组:空白对照组,MLT组。每组均加入对氯苯丙氨酸(parachlorophenylalanine,PCPA),以抑制淋巴细胞MLT的内源性合成,并在植物凝集素(PHA)刺激下培养。于20h后取部分培养细胞用流式细胞仪测定CD4+CD69+细胞百分比和平均荧光强度;72h取剩余部分细胞用流式细胞仪测定CD4+CD25+和CD4+HLA.DR+细胞百分比和平均荧光强度。经方差分析检验统计分析数据,观察MLT对CD4+T细胞活化的影响。结果在PCPA抑制淋巴细胞内源性MLT合成和PHA的刺激下,外源性MLT能有效增强CD4+T细胞的活化,使得CD25、CD69、HLA.DR的CD4+T细胞表达的百分比显著增加(P〈0.05);但间接用于表示单个细胞表面分子数量的平均荧光强度并不明显增加(P〉0.05)。结论MLT能有效地增强PHA对成人CD4+T细胞的活化作用,但并不能增加单个CD4+T细胞表面CD25、CD69、HLA.DR分子表达的量。
Objective To investigate the role of melatonin (MLT) and its membrane receptor effecting the activating process of CD4+ T cell in adults. Methods 27 cases of isolated peripheral blood mononuclear cells from normal adults, each specimen was divided into 2 groups, first group is the control, second group was added MLT. With the inhibition of MLT synthesis by the parachlorophenylalanin (PCPA) in lymphocytes and PHA's stimulation, these peripheral blood mononuclear cells were cultured. After 20 hours, we took some cultured cells to detect CD4+CD69+ cell percentage and mean fluorescence index by flow cytometry, and 72 hours later analysed the remainder of the cells by flow cytometry to know CD4+CD25+ and CD4+HLA-DA+ cell percentage and the mean fluorescence intensity. By paired T test analyse the data, we observed the role of MLT and its receptor antagonist on CD25, CD69, HLA-DR expression. Results The inhibition of MLT synthesis by the PCPA and the lymphocytes were stimulated by PHA, MLT can effectively stimulate CD4+ T cells and increase the percentages of CD4+CD25+, CD4+CD69+, CD4+HLA-DR+ T cells (P 〈 0.05); but cannot enhance the mean fluorescence intensity (P 〉 0.05). Conclusion These findings suggest that MLT can effectively enhance the CD4+ T cells' activation by PHA in adults, but cannot raise the amount of CD25, CD69, HLA-DR molecule expressing on every CD4+ T cell surface.
