摘要
目的:研究以Aptamer功能化水凝胶实现卵巢癌循环肿瘤细胞的捕捉与无损伤释放。方法:Aptamer功能化水凝胶是一种新型的生物材料,采用DNA具有可逆性结合的特性,用一段DNA Aptamer捕获目标分子,形成水凝胶体系,实现对目标分子的分离。结果:二级结构的寡核苷酸分子可以有效地让初级结构的寡核苷酸分子与配体分离并且恢复配体活性,成功实现卵巢癌循环肿瘤细胞捕捉与无损伤释放。结论:Aptamer水凝胶可以控制细胞间相互作用,实现卵巢癌循环肿瘤细胞无损伤捕捉与释放。
Objective: To study the capture and release of circulating tumor cells in patients with-ovarian cancer through hydrogel functionalize with Aptamer. Methods: Hydrogel functionalize with Aptamer is a kind of new biological material.Based on the reversiblly binding properties of DNA fragments, target molecules were captured and isolated.Results:Secondary structure of oligonucleotide molecules could effectively have the primary structure isolated from ligand and so the ligand regained activity.Circulating tumor cells in patients with ovarian cancer were captured and released successfully without damage. Conclusion.. Circulating tumor cells in patients with ovarian cancer can be captured and released without damage through hydrogel functionalize with Aptamer.
