超高效液相色谱法测定糜子中4种B族维生素 被引量:2

Simultaneous Determination of Four Water-soluble B Vitamins in Panicum miliaceum L. by Ultra Performance Liquid Chromatography

摘要 建立同时测定糜子中4种B族维生素的超高效液相色谱法。糜子样品用0.01 mol/L盐酸超声加热溶解,醋酸锌沉淀蛋白后提取滤液,采用WatersHCLASS超高效液相色谱仪,以C_(18)柱为固定相,乙腈-1%甲酸溶液(pH=2.63)为流动相,流速0.25 mL/min梯度洗脱,利用紫外检测器在波长275 nm处进行检测,分析时间为11 min。结果表明,维生素B_1,B_2,B_6,B_9这4种维生素线性关系良好,R^2均在0.999以上,相对标准偏差(RSD)为0.42%~1.56%,回收率范围为91.70%~101.02%。该法快速简便,精密度好,结果准确可靠,适用于同时测定糜子中水溶性维生素的含量,可对糜子营养品质进行评价,鉴选优良种质。 To establish simultaneous determination of 4 kinds of water-soluble vitamin B in Panicum miliaceum L.used by ultra performance liquid chromatography(UPLC),samples were dissolved by ultrasonic heating with 0.01 mol/L of hydrochloric acid and filtrate was extracted after protein was precipitated by zinc acetate.C_(18) was stationary phase and acetonitrile-1%formic acid solution was moving phase.Gradient elute was done at the rate of 0.25 mL/min.Detection was done at 275 nm of wave length used by ultraviolet detector and analysis time was 11 min.The result showed that linear relations of vitamin B_1,vitamin B_2,vitamin B_6,and vitamin B_9 were good.Average value of R-2 was over 0.999.Relative standard deviation was 0.42%-1.56%,while recovery rate was 91.70%-101.02%.The method is fast,convenient,accurate and reliable,can be applied in determining content of water soluble vitamin and appraising and selecting excellent Panicum miliaceum L.germplasmresources.
