摘要
SCD1是位于内质网上的一类催化酶,在肝脏的脂质代谢及鹅肥肝形成中发挥重要作用。试验通过荧光定量PCR技术对填饲不同阶段(填饲7、14和19 d)朗德鹅肝脏中SCD1基因的表达进行了研究。结果发现:SCD1基因在填饲各阶段肝脏中表达量均高于对照组,且填饲19 d时,填饲组表达量极显著高于对照组(P<0.01);通过测定填饲组各时期肝脏中棕榈油酸、油酸含量以及血液中甘油三酯和胆固醇含量发现,SCD1基因表达量与油酸、胆固醇、VLDL以及甘油三酯含量均呈显著正相关(P<0.05);通过生物学软件预测SCD1的靶向miRNA并利用双荧光素酶报告系统在CHO细胞系中验证所筛选的3个miRNA,结果显示miR30b与miR-30a-5p为鹅SCD1基因可能的靶向miRNA,进一步研究发现miR-30b和miR-30a-5p在填饲组表达量均显著低于对照组(P<0.05),表明miR-30b和miR-30a-5p可能通过对SCD1基因的调节作用影响鹅肝脏脂代谢过程。
SCD1 is a kind of catalysis enzyme which is located in the endoplasmic reticulum. It plays a pivotal role in lipid metabolism and goose fatty liver formation. In this study, the expression profiles of SCD1 gene in goose liver at different stages of overfeeding (7, 14 and 19 days) were determined by qRT-PCR. The results showed that the expression of SCD! gene was higher in overfeeding group at all stages of overfeeding,and at day 19 of overfeeding,the SCD1 expression of overfeeding group was significantly higher than control group (P〈0.01). The levels of palmitoleic acid, oleic acid, triglycerides and cholesterol in liver at every stage of overfeeding period were also detected, and the correlation between these indexes with SCD1 gene expression was analyzed. The expression of SCD1 was significantly positively correlated with the concentrations of oleic acid, cholesterol, VLDL and triglycerides (P〈0.05). The miRNAs that may regulate goose SCD1 were predicted by bioinformatic software, and the candidate miRNAs were validated in CHO cells using dual luciferase reporter system. It turned out that miR-30b and miR-30a-Sp could regulate the expression of SCD1 gene. It was also found that the expression of miR-30b and miR-30a-5p were lower in the over-feeding group, which revealed that miR-30b and miR-30a-5p might affect the lipid metabolism of goose by targeting SCD1 gene.
