摘要
为确诊海南省某鸡场疑似心包积液综合征病例,于2016年5月从海南省某鸡场送检的病鸡中采集病料(肝脏、心脏),研磨,通过卵黄囊接种7日龄SPF鸡胚,并进行鸡胚病变特征观察、禽腺病毒PCR特异性检测、基因测序和Blast序列比对。结果表明,接种鸡胚后5d开始死胚,剖检可见胚体发育受阻,羽毛稀少,体表潮红,鸡胚皮肤表面有出血点;在病料研磨液、尿囊液及胚体肝脏中均检测到大小约为884bp目的片段,将分离病毒命名为HN1605;hexon基因Blast序列比对及遗传进化分析结果表明,分离毒株与2015年山东和湖北报道的Ⅰ亚群腺病毒血清4型(FAV-4)SDSX、SDSX-15、SDNY2-15、HB1510和HBLF-15分离株亲缘关系最近。本研究表明,此次心包积液综合征病原是Ⅰ亚群禽腺病毒血清4型病毒,该病毒依然危害着我国鸡群,疫情不断扩大,并呈现由我国北方地区向南方地区扩散的趋势。
In this research,a fowl adenovirus serotype 4 strain named GD1605 was isolated and identlhed from livers and hearts of sick chickens in a farm of Hainan province in China, May 20t6,based on SPF chicken embryo culture, chicken embryo pathological lesions, PCR assay, BLAST and sequence analysis. The results showed that each of inoculated chicken embryo appeared to be dead 5 d to 7d post inoculation and the death embryoes were observed as arrested development,feather scarce and skin hemorrhage.An a- vian adenovirus-specific PCR product (884 bp) was amplified and sequenced.The genetic evolution analysis results showed that the virus shared high identity with FAV-4 in group I .The genetic evolution analysis also indicated that the virus was closely related to the FAV-4(group I) strains of SDSX,SDSX-15,SDNY2- 15, HB1510 and HBLF-15.These results confirmed that the isolated virus is a member of fowl adenovirus serotype 4 in group I ,and causes hydropericardium-hepatitis syndrome.This virus still endangering chick- en flocks in China, the epidemic expands unceasingly, and display the trend of epidemic from north to south regions in China.
