摘要
目的探讨磷脂酰肌醇-3激酶(PI3K)抑制剂对高糖环境下肾小球足细胞上皮间质转分化(EMT)的影响。方法体外培养原代大鼠肾小球足细胞,分为NG组(葡萄糖5mmol/L)、MG组(甘露醇30 mmol/L)、HG组(葡萄糖30 mmol/L)、HG+LY294002组(葡萄糖30 mmol/L+LY294002 2mg/L),培养48h后检测α-平滑肌肌动蛋白(α-SMA)和E-cadherin蛋白的表达情况。结果 HG组足细胞足突消失及失去细胞间连接。与NG组比较,HG组α-SMA蛋白表达增多,E-cadherin蛋白表达减少(P<0.05);与HG组相比,HG+LY294002组α-SMA蛋白表达减少,E-cadherin蛋白表达增加(P<0.05)。结论高糖导致足细胞损伤发生EMT,PI3K抑制剂可部分阻断EMT的发生。
Objective To investigate the effect of the PI3K inhibitor on epithelial mesenchymal transdifferentiation(EMT) of glomerulus podocytes in high glucose environment. Methods Primary cultured rat glomerulus podocyte cells in vitro were further cultured with glucose 5 mmol/L(group NG) ,mannitol 30 mmol/L(group MG), glucose 30 mmol/L(group HG), or glucose 30 mmol/L plus LY294002 2 mg/L(group HG+LY294002) for 48 hours. The protein expression levels of α-SMA and E-cadherin were detected after cultured for 48 hours. Results The foot process of podocyte cells disappeared and intercellular connection lost in group HG. Compared with group NG, the protein expression of α-SMA was increased, and that of E-cadherin was reduced in group HG(P〈0. 05). Compared with group HG, the protein expression of α-SMA was decreased and that of E-cadherin was increased in group HG+ LY294002 (P〈0.05). Conclusion High glucose can induce EMT of the podocytes, which can be blocked partly by phosphatidy linositol 3-kinase inhibitor.
