摘要
目的:探讨补气消癜汤对免疫性血小板减少症模型(ITP)小鼠的白细胞介素系列因子及肿瘤坏死因子-α(TNF-α)的影响,旨在为临床治疗提供依据。方法:采用注射豚鼠抗小鼠血小板血清(GP-APS)方法建立ITP小鼠,随机将72只小鼠分为6组:空白组、模型组、强的松组、补气消癜汤组(低、中、高剂量),观察造模前、给药前、给药后小鼠外周血血小板计数(PLT);给药后,观察小鼠的脾脏系数、胸腺系数、骨髓巨核细胞数(MN)、脾脏MN,采用ELISA法测定小鼠血清白介素-2(IL-2)、IL-6、IL-10及肿瘤坏死因子-α(TNF-α)、血小板生成素(TPO)含量的变化。结果:1给药前,与空白组对比,模型组、强的松组、补气消癜汤各剂量组的PLT均出现明显下降(P<0.01);给药后,各组小鼠的PLT较造模前减少,较给药前增多,差异显著(P<0.01);与模型组对比,补气消癜汤各剂量组的PLT和TPO明显增多(P<0.01)。2与空白组对比,模型组的脾脏系数、胸腺系数、脾脏MN、骨髓MN均明显增多(P<0.01),与模型组对比,补气消癜汤各剂量组的脾脏系数、胸腺系数、脾脏MN、骨髓MN明显减少(P<0.01或P<0.05),与强的松组对比,补气消癜汤高剂量组的脾脏系数、胸腺系数、脾脏MN明显减少(P<0.05)。3与空白组对比,模型组的IL-2、IL-6及TNF-α升高,IL-10降低,差异显著(P<0.01);与模型组对比,强的松组、补气消癜汤各剂量组的IL-2、IL-6及TNF-α降低,IL-10升高,差异显著(P<0.01);与强的松组对比,补气消癜汤高剂量组的IL-2及TNF-α降低,IL-10升高,差异显著(P<0.01或P<0.05)。结论:补气消癜汤可通过调节ITP小鼠体内IL-2、IL-6、IL-10及TNF-α分泌,促进脾脏和骨髓的巨核细胞分化,增加ITP模型小鼠外周血小板数。
Objective : To explore the influence of interleukins factors and TNF -α in mice model of immune thrombo- cytopenia by Buqi Xiaodian Decoction and provide the basis for clinical treatment. Methods : By using injections of Guinea pig anti platelet serum into mice to establish ITP model,72 mice were randomly divided into 6 groups: blank group, model group, prednisone group, Buqi Xiaodian Decoction (low dose, middle dose and high dose) groups. The peripheric platelet counts before model, before medicine and after medicine were observed. After treatment, thymus coefficient, spleen coefficient,bone marrow megakaryocyte number and spleen megakaryocyte number were observed. The change of serum inter- leukin- 2 ,IL- 6 ,IL- 10 and tumor necrosis factor-α and platelet hormone in mice were detected by ELISA method. Results: (1)Before treatment, compared with blank group, PLT in model group, prednisone group and Buqi Xiaodian Decoction each dose group were significantly lower (P 〈 0. 01 ). After treatment, the PLT reduced compared with that before modeling and increased compared with that before medicine. There was significant difference (P 〈 0.01 ). Compared with model group, PLT and TPO in Buqi Xiaodian Decoction each dose group were obviously increased (P 〈 0.01 ). (:2)Compared with blank group, thymus coefficient, spleen coefficient, spleen MN and bone MN in model group were significantly increased (P 〈 0. 01 ). Compared with model group, thymus coefficient, spleen coefficient, spleen MN and bone MN in Buqi Xiaodian Decoction each dose group decreased significantly ( P 〈 0. 01 or P 〈 0.05 ). Compared with prednisone group,thymus coefficient, spleen coefficient and spleen MN in Buqi Xiaodian Decoction high dose group decreased signifi- cantly ( P 〈 0. 05 ).
