上皮-间充质转化相关分子与三阴性乳腺癌不良预后的相关性 被引量:5

Correlation between epithelial- mesenchymal transition related markers and poor clinical outcome of triple- negative breast cancer

摘要 目的检测三阴性乳腺癌(TNBC)患者的上皮-间充质转化(EMT)标记分子(波形蛋白、平滑肌肌动蛋白、骨粘连蛋白、N-钙黏素以及E-钙黏素缺失)和EMT诱导分子锌指E-盒结合同源异形盒-1 (ZEB1)的表达,探讨它们的表达与TNBC的临床病理特征之间的联系。方法对173例TNBC乳腺癌样本上述EMT相关分子免疫组织化学检测,并将这些分子的表达与疾病的临床病理特征进行相关分析。结果波形蛋白在基底样TNBC(87/154,56.5%)中的表达多于非基底样TNBC(3/17,17.6%)。ZEB1的表达只与平滑肌肌动蛋白的表达有相关性(P=0.002)。生存分析显示波形蛋白(P=0.515)、平滑肌肌动蛋白(P=0.739)、骨粘连蛋白(P=0.094)、N-钙黏素(P=0.509)和E-钙黏素(P=0.925)与患者的预后没有明显相关,而ZEB1可作为不良无病生存期的独立预后因素(P=0.007)。结论EMT相关分子可作为TNBC分化型亚组的独特标记。 Objective To investigate the expression of epithelial-mesenchymal transition (EMT) related molecular markers (vimentin, smooth muscle actin, osteonectin, N-cadherin, and E-cadherin loss) and EMT inducer zinc-finger E-box binding homeobox 1 (ZEB1) in triple-negative breast cancer (TNBC) samples and inquire the correlation between their expression and clinicopathological features of the tumors.Methods We analyzed immunohistochemical expression of EMT-related markers mentioned above in 173 TNBC samples and correlation between their expression with clinicopathological features of the tumors, including clinical outcome.Results The expression levels of vimentin were more frequent in basal-like TNBC (87/154, 56.5%) than in non-basal-like TNBC (3/17, 17.6%). The ZEB1 expression was correlated only with the expression of smooth muscle actin (P=0.002). The survival analyses revealed that vimentin (P=0.515), smooth muscle actin (P=0.739), osteonectin (P=0.094), E-cadherin (P=0.925) and N-cadherin (P=0.509) were not associated with patients’ clinical outcomes. However, the ZEB1 expression was found to be an independent prognostic factor for poor disease-free survival.Conclusion These findings indicate that the expression of EMT-related markers in TNBC can be a signature of a certain subgroup of TNBC.
