氢对内毒素致巨噬细胞氧化损伤时Nrf2表达的影响 被引量:1

Effect of hydrogen on expression of nuclear factor E2-related factor 2 during endotoxin-induced oxi- dative injury to macrophages

摘要 目的评价氢对内毒素致巨噬细胞氧化损伤时核因子E2相关因子2(Nrf2)表达的影响。 方法正常培养的RAW264.7细胞,采用随机数字表法分为4组(n=36):对照组(C组)、富氢液组(H组)、内毒素组(E组)和内毒素+富氢液组(EH组)。E组和EH组加入内毒素,终浓度1 μg/ml,H组和EH组加入富氢液(氢气浓度0.06 mmol/L),于富氢液孵育6、24和48 h时收集细胞,检测ROS活性,收集细胞提取蛋白,采用Western blot法检测SOD、过氧化氢酶(CAT)活性和Nrf2的表达。 结果与C组比较,E组和EH组ROS活性升高,SOD和CAT活性降低,巨噬细胞Nrf2表达上调(P〈0.05),H组上述指标差异无统计学意义(P〉0.05);与E组比较,EH组ROS活性降低,SOD和CAT活性升高,巨噬细胞Nrf2表达上调(P〈0.05)。 结论氢可减轻内毒素致巨噬细胞氧化损伤,其机制与上调Nrf2表达有关。 Objective To evaluate the effect of hydrogen on the expression of nuclear factor E2- related factor 2 ( Nrf2 ) during endotoxin-indueed oxidative injury to macrophages. Methods Normally cuhured Raw264.7 eells were divided into 4 groups (n= 36 each) using a random number table: control group (group C), hydrogen-rich saline group (group H), endotoxin group (group E) and endotoxin plus hydrogen-rich saline group (group EH). In E and EH groups, endotoxin was added with the final concentration of 1 txg/ml. Hydrogen-rich saline (hydrogen concentration 0.06 mmol/l,) was added in H and Eli groups. All the cells were incubated for 48 h. At 6, 24 and 48 h of incubation, cells were collect- ed to detect the activity of reactive oxygen species (ROS) , and (:ells were collected and proteins extracted for determination of superoxide dismutase (SOD) and catalase (CAT) activities and Nrt2 expression by Western blot. Results Compared with group C, the ROS activity was significantly increased, the levels of SOl) and CAT were significantly decreased, and the expression of Nrf2 was significantly up-regulated in E and EH groups (P〈O.05), and no significant change was tound in the parameters mentioned above in group H (P〉0.05). Compared with group E, the ROS activity was significantly decreased, the levels of SOD and CAT were significantly increased, and the expression of Nrf2 was signifieantly up-regulated in group EH (P〈0.05). Conclusion Hydrogen can attenuate endotuxin-induced oxidative injury to macro- phages, and the mechanism may be related to up-regulated expression of Nrf2.
